毫针针刺足三里对失神经肌萎缩大鼠穴区电阻、温度和能量代谢变化的影响

Effect of Acupoint Resistance,Temperature and Energy Metabolismof Rats with Denervation Muscular Atrophy after Acupuncture at Zusanli Point

摘要 目的研究大鼠胫前肌失神经萎缩对毫针针刺腧穴穴区电阻、温度及能量代谢的影响,为不同机能状态影响毫针针刺作用差异的生物医学机制提供实验依据。方法将32只清洁级SD大鼠随机分为正常组和肌萎缩组,每组大鼠再配对分入针刺组和非针刺组2个亚组。使用A型肉毒素肌肉注射建立大鼠胫前肌化学性失神经支配肌萎缩模型,针刺组针刺“足三里”穴,捻转频率60次/min,行针5 min,留针10 min,共操作3轮后出针,每日操作1次,连续3 d。针刺结束后,测量穴区电阻和温度,切取以穴点为圆心,直径0.5 cm,深度0.5 cm区域的组织块,采用酶联免疫法检测组织中ATP、ADP的含量并计算ATP/ADP。结果针刺正常组大鼠可以降低腧穴的电阻、温度,提高穴区ATP/ADP,差异均有统计学意义(P<0.01);针刺肌萎缩组大鼠能降低穴区电阻、温度,差异均具有统计学意义(P<0.01),且电阻降低幅度显著大于正常组(P<0.01);针刺肌萎缩组大鼠不能提高穴区ATP/ADP,与正常针刺组比较,差异具有显著统计学意义(P<0.01)。结论肌萎缩状态下毫针针刺激活腧穴的作用与正常机体状态存在不同,肌萎缩状态可明显减弱针刺对腧穴ATP/ADP的调节幅度,针刺对肌萎缩的治疗作用可能主要通过增强腧穴电敏特性达成。 Objective:To study the effects of denervation atrophy of rat anterior tibial muscle on acupoint resistance,temperature and energy metabolism in the acupoints of acupuncture needles,and to provide experimental evidence for the biomedical mechanism of affecting the effect difference of acupuncture needles in different functional states.Methods:32 clean SD rats were randomly divided into a normal group and a muscular atrophy group,and each group was paired into two subgroups,acupuncture group and non-acupuncture group.The rat model of chemical denervation muscle atrophy of tibialis anterior muscle was established by intramuscular injection of botulinum toxin type A.The acupuncture group was injected at Zusanli point,the twisting frequency was 60 times/min,the needle was inserted for 5 min,the needle was retained for 10 min,and the needle was extracted after 3 rounds of operation,once a day for 3 consecutive days.After the acupuncture,the resistance and temperature of the acupoint area were measured,and tissue blocks with the acupoint as the center of the circle with a diameter of 0.5cm and a depth of 0.5cm were cut.The contents of ATP and ADP in the tissues were detected and ATP/ADP was calculated by ELISA.Results:The acupuncture in normal group could reduce the resistance and temperature of the acupoints and increase ATP/ADP in the acupoints,and the differences were statistically significant(P<0.01).The acupuncture in muscular atrophy group could reduce the resistance and temperature in the acupoint area,and the difference was statistically significant(P<0.01),and the reduction of resistance was significantly greater than that in normal group(P<0.01).The acupuncture in muscular atrophy group could not improve ATP/ADP in the acupoint area,and the difference was statistically significant compared with that of the normal acupuncture group(P<0.01).Conclusion:The effect of acupuncture activation on acupoints under the condition of muscular atrophy is different from that of normal body condition.The condition of muscular atrophy can significantly weaken the adjustment amplitude of acupuncture on ATP/ADP of acupoints.The therapeutic effect of acupuncture on muscular atrophy may be mainly achieved through enhancing the electrical sensitivity characteristics of acupoints.
