应用LSDV-ELISA抗体检测试剂盒检测羊痘疫苗免疫牛血清抗体水平

Use the ELISA Kit for Detection of Antibodies Against Lumpy Skin Disease Virus to Detect the Antibody Level of Bovine Serum Immunized with Goat Pox Vaccine

目前国内尚无商品化的牛结节性皮肤病疫苗,对于该病的防控,我国采用羊痘疫苗进行紧急免疫。为了验证牛结节性皮肤病病毒ELISA抗体检测试剂盒对羊痘疫苗免疫血清抗体水平检测的适用性,本研究采用实验室试制的牛结节性皮肤病病毒ELISA抗体检测试剂盒和病毒中和试验对羊痘病毒活疫苗免疫血清、在研牛结节性皮肤病灭活疫苗(山羊痘AV41株)免疫血清进行抗体水平评价。结果显示,对于羊痘活疫苗免疫血清,ELISA方法和病毒中和试验方法对免疫15d抗体阳性率分别为84.4%和75.6%,免疫30d抗体阳性率分别为100%和95.6%,免疫60d抗体阳性率分别为97.8%和91.1%;与病毒中和试验符合率为86.1%。对于牛结节性皮肤病灭活疫苗(山羊痘AV41株)免疫血清,ELISA方法和病毒中和试验方法对一免28d抗体阳性率分别为64%和56%,二免28d抗体阳性率分别为94%和88%,二免42d阳性率均为100%;与病毒中和试验符合率为86.5%。试验证明,实验室试制的牛结节性皮肤病病毒ELISA抗体检测试剂盒与病毒中和试验符合率较高,检测结果较为一致,且本试剂盒敏感性高于病毒中和试验。因此本试剂盒可用于牛结节性皮肤病抗体水平检测。

At present,there is no vaccine against lumpy skin disease in China.For the prevention and control of the disease,sheep pox vaccine is used for emergency immunization.In order to verify the ELISA kit for detection of antibodies against lumpy skin disease virus for detecting sheep pox vaccine immune serum antibody level ofapplicability,this research adopts the laboratory produts for the ELISA kit for detection of antibodies against lumpy skin disease virus of goatpox virus vaccine immune serum,lumpy skin disease inactivated vaccine(goat pox AV41 strain)immune serum antibody level evaluation.The results showed that the antibody positive rate of ELISA kit and virus neutralization test were 84.4%and 75.6%for 15 days,100%and 95.6%for 30 days and 97.8%and 91.1%for 60 days immunized against sheep pox vaccine,and the coincidence rate with virus neutralization test was 86.1%.For lumpy skin disease inactivated vaccine immune serum,the positive rates of ELISA and virus neutralization test were 64%and 56%after 28 days of first immunization,94%and 88%after 28 days of second immunization,100%and 100%after 42 days of second immunization,the coincidence rate with virus neutralization test was 86.5%.The experimental results showed that the ELISA antibody test kit had a high coincidence rate with the virus neutralization test,and the detection results were consistent,and the sensitivity of the kit was higher than that of the virus neutralization test.Therefore,this kit can be used to detect antibody levels in lumpy skin disease.
