摘要
为了探究禽传染性支气管炎病毒非编码RNA对芳烃受体(AhR)及其信号通路相关基因表达的调控作用,将rIBV和rIBV-C27107G分别感染H1299细胞16 h后,用qRT-PCR检测AhR及其通路相关基因的表达水平,并用Western blot检测AhR蛋白的丰度。结果显示,不产生ncRNA的IBV感染可激活AhR信号通路,诱导H1299细胞产生抗病毒反应,而IBV编码的ncRNA抑制AhR信号通路的激活,从而抑制细胞因子IL-6、IL-8和IL-12β的上调表达,利于病毒的复制。
In order to explore the regulatory effect of avian infectious bronchitis virus non-coding RNA on aryl hydrocarbon receptor(AhR)and related genes in signaling pathway,H1299 cells were infected with rIBV and rIBV-C27107G for 16 h.The expressions of these genes in the IBV-infected H1299 cells were detected by qRT-PCR and the expression level of AhR protein was detected by Western blot.The results show that IBV infection without ncRNA can activate the AhR signaling pathway and induce an antiviral response in H1299 cells,while IBV-encoded ncRNA can inhibite the activation of the AhR signaling pathway,thus inhibiting the up-regulation of cytokines IL-6,IL-8 and IL-12β,which is conducive to viral replication.
