肝细胞肝癌组织中TFAM、PDLIM1表达及与患者临床病理特征和预后的关系 被引量:1

Expression of TFAM and PDLIM1 in hepatocellular carcinoma and their relationships with clinicopathological characteristics and prognosis

摘要 目的探究肝细胞肝癌(HCC)组织中线粒体转录因子A(TFAM)和PDZ-Lim结构域蛋白1(PDLIM1)表达及与患者临床病理特征和预后的关系。方法采用免疫组化法检测103例HCC患者癌和癌旁组织中TFAM和PDLIM1蛋白表达。Spearman秩相关分析TFAM和PDLIM1蛋白表达的相关性。比较不同临床病理特征HCC患者TFAM、PDLIM1蛋白表达差异。应用Kaplan-Meier法分析TFAM、PDLIM1蛋白表达与HCC患者预后的关系。利用单因素及多因素Cox比例回归模型分析影响HCC患者预后的因素。结果HCC癌组织中TFAM和PDLIM1蛋白表达阳性率分别为21.36%(22/103)、29.13%(30/103),癌旁组织中TFAM和PDLIM1蛋白表达阳性率分别为76.70%(79/103)、78.64%(81/103),癌组织中TFAM和PDLIM1蛋白表达阳性率低于癌旁组织(χ2分别为63.111、50.811,P均<0.05)。HCC癌组织中TFAM与PDLIM1表达呈正相关(r=0.801,P<0.05)。不同肿瘤最大径、肿瘤TNM分期HCC癌组织中TFAM和PDLIM1蛋白表达阳性率差异有统计学意义(P均<0.05)。TFAM阴性表达者3年累积生存率低于TFAM阳性表达者(P<0.05)。PDLIM1阴性表达者3年累积生存率低于PDLIM1阳性表达者(P<0.05)。肿瘤TNM分期Ⅲ期、肿瘤最大径>3 cm、TFAM阴性表达、PDLIM1阴性表达是影响HCC患者预后的独立危险因素(P均<0.05)。结论HCC癌组织中TFAM、PDLIM1表达降低,二者表达与肿瘤最大径及肿瘤分期有关;TFAM、PDLIM1可能成为新的HCC预后相关肿瘤标志物。 Objective To investigate the expression levels of mitochondrial transcription factor A(TFAM)and PDZLim domain protein 1(PDZ and LIM domain 1,PDLIM1)in hepatocellular carcinoma(HCC)tissues and their relationships with the clinicopathological characteristics and prognosis.Methods The expression levels of TFAM and PDLIM1 proteins in 103 HCC patients´cancer and adjacent tissues were detected by immunohistochemistry.Spearman rank correlation was used to analyze the correlation between TFAM and PDLIM1 protein expression.The expression difference of TFAM and PDLIM1 proteins in HCC patients with different clinicopathological characteristics was compared.Kaplan-Meier method was used to analyze the relationships between the expression levels of TFAM,PDLIM1 proteins and survival prognosis of HCC patients after surgery.Single-factor and multi-factor Cox regression models were used to analyze the factors affecting the survival prognosis of HCC patients after surgery.Results The positive expression rates of TFAM and PDLIM1 proteins were 21.36%(22/103)and 29.13%(30/103),respectively,in the HCC tissues,and 76.70%(79/103)and 78.64%(81/103),respectively,in the adjacent tissues;the positive expression rates of TFAM and PDLIM1 proteins in the HCC tissues were lower than those in the adjacent tissues(χ2=63.111,50.811,both P<0.05).There was a significant positive correlation between TFAM and PDLIM1 expression in HCC tissues(Rs=0.801,P<0.05).There were statistically significant differences in the positive expression rates of TFAM and PDLIM1 proteins among HCC tissues of different tumor maximum diameters and tumor stages(all P<0.05).The 3-year cumulative survival rates of patients with negative TFAM expression was significantly lower than that in patients with positive TFAM expression(P<0.05).The 3-year cumulative survival rate of patients with negative PDLIM1 expression was significantly lower than that of patients with positive PDLIM1 expression(P<0.05).Tumor TNM stageⅢ,tumor maximum diameter>3 cm,negative expression of TFAM,and negative expression of PDLIM1 were independent risk factors for prognosis of HCC patients(all P<0.05).Conclusion The expression levels of TFAM and PDLIM1 decrease in HCC tissues,their expression is related to the max-imum diameter and tumor staging of the tumor,and TFAM and PDLIM1 may be new tumor markers related to prognosis of HCC.
