长链非编码RNA RAB11B-AS1在胶质瘤中的表达及预后价值研究

Expression and prognostic value of lncRNA RAB11B-AS1 in glioma

目的:检测长链非编码RNA(lncRNA)RAB11B-AS1(RAB11B-AS1)在脑胶质瘤中的表达特征,分析其在不同临床病理特征中的表达差别,探讨其预后价值。方法:应用生信分析数据库(GEPIA)对RAB11B-AS1的表达特征(163例胶质瘤、207例正常脑组织)进行预测。选择68例确诊为脑胶质瘤并行手术治疗的患者作为研究对象,留取术后肿瘤组织,选择68例因脑出血行手术治疗后留取的边缘正常脑组织作为对照。选择人脑胶质瘤细胞株U87和T98G,选择人脑正常胶质细胞株HEB。应用实时荧光定量PCR法(qRT-PCR)检测RAB11B-AS1的表达,应用免疫组化EnVision检测增殖细胞核抗原(PCNA)的表达。结果:GEPIA预测到RAB11B-AS1在胶质瘤中的表达可能升高。脑胶质瘤术后组织中RAB11B-AS1的表达明显高于正常脑组织(P=0.0001)。胶质瘤中RAB11B-AS1表达在不同肿瘤最大径、WHO分级、有无坏死方面比较差异有统计学意义(均P<0.05)。胶质瘤中RAB11B-AS1与PCNA具有正相关性(r=0.62,P=0.0341)。生存分析显示RAB11B-AS1的表达与术后生存时间有关(P=0.041)。胶质瘤细胞株U87和T98G中RAB11B-AS1的表达均明显高于HEB(均P<0.05)。结论:脑胶质瘤组织中RAB11B-AS1的表达升高,与PCNA的表达具有正相关性,术后检测RAB11B-AS1的表达对判断预后可能有一定价值。

Objective:To detect the expression characteristics of lncRNA RAB11B-AS1(RAB11B-AS1)in glioma,analyze the differences on different clinicopathological features,and explore its prognostic value.Methods:The expression characteristics of RAB11B-AS1(163 cases of glioma and 207 cases of normal brain tissue)were predicted by GEPIA.Sixty-eight patients diagnosed with glioma and treated with surgery were selected as study subjects,and postoperative tumor tissues were retained.Sixty-eight cases of marginal normal brain tissue collected after surgical treatment of cerebral hemorrhage were selected as controls.Human glioma cell lines U87 and T98G and human normal glial cell line HEB were selected.The expression of RAB11B-AS1 was detected by qRT-PCR,and proliferating cell nuclear antigen(PCNA)was detected by immunohistochemistry EnVision method.Results:GEPIA observations predicted a possible elevated expression of RAB11B-AS1 in gliomas.The expression of RAB11B-AS1 was significantly higher in glioma tissue than that of normal brain tissue(P=0.0001).Expression of RAB11B-AS1 was different in the maximum diameter and WHO grading,with or without necrosis of glioma(P<0.05).Positive correlation was found between RAB11B-AS1 and PCNA(r=0.62,P=0.0341).RAB11B-AS1 expression was associated with postoperative survival time by survival analysis(P=0.041).The expression of RAB11B-AS1 was significantly higher in both glioma cell lines U87 and T98G than that in HEB(P<0.05).Conclusion:The expression of RAB11B-AS1 was increased in glioma,which was positively correlated with PCNA.Postoperative detection of RAB11B-AS1 expression may have certain value in predicting prognosis.
