GLUT基因表达对异色瓢虫幼虫能量代谢的影响

摘要 异色瓢虫Harmoniaaxyridis是一种重要的捕食性瓢虫,可捕食多种蚜虫及其他害虫。目前,已被世界各地广泛用于农业害虫的生物防治。为实现短时间低成本快速扩繁异色瓢虫,人们对异色瓢虫人工饲料的配方进行了大量研究,使其更好地完成瓢虫的生长发育。实验室前期筛选出添加海藻糖和葡萄糖的人工饲料,能增加幼虫体质量、缩短瓢虫产卵前期、提升产卵量和卵孵化率等,有助于瓢虫的生长发育和繁殖。本文以饲喂蚜虫组为对照组,对2个葡萄糖转运蛋白基因(GLUT1和GLUT4)在异色瓢虫发育各阶段的表达情况进行了研究,并通过RNA干扰技术,进一步探究了注射ds GLUT1或ds GLUT4后异色瓢虫体内能量代谢相关基因的表达情况和酶活性变化,从而证明该基因与异色瓢虫幼虫能量代谢的调节功能密切相关。结果表明:注射ds GLUT1和ds GLUT4显著影响了TPS、TRE2、TRE2-like等基因的表达,同时发现在干扰GLUT1和GLUT4在幼虫体内的表达后,糖原、葡萄糖、海藻糖等糖类物质含量均有所下降。表明GLUT1和GLUT4在异色瓢虫幼虫的繁殖中起到一定的调控作用。研究结果有助于为未来进一步研究关键基因表达与异色瓢虫幼虫能量代谢之间的关系提供理论支持,为合理地利用异色瓢虫进行生物防治提供技术指导和参考依据。 Harmonia axyridis is an important predatory ladybug that preys on a variety of aphids and other pests.At present,it has been widely used in the biological control of agricultural pests all over the world.In order to achieve rapid propagation of ladybugs at low cost in a short period of time,a lot of research has been done on the formula of artificial diet for ladybugs,so that they can complete growth and development.The artificial diet supplemented with trehalose and glucose was screened out in the laboratory,which can increase ladybug larval body weight,shorten the pre-oviposition period,and increase fecundity and the egg hatching rate,and benefit the growth,development and reproduction of the ladybugs.With ladybugs feeding aphid as the control group,the expression of two glucose transporter genes(GLUT1 and GLUT4) at various developmental stages of H.axyridis was analyzed,and the RNA interference technology was used to further explore the expression of dsGLUT1 or dsGLUT4 after injection.The expression and enzymatic activity of energy metabolism-related genes in H.axyridis proved that this gene is closely related to the regulation function of energy metabolism in H.axyridis larvae.The results showed that injection of dsGLUT1 and dsGLUT4 significantly affected the expression of TPS,TRE2,TRE2-like and other genes,and it was found that the contents of glycogen,glucose,trehalose and other carbohydrates decreased after interfering with the expression of GLUT1 and GLUT4 in larvae.It indicates that GLUT1 and GLUT4 play a certain regulatory role in the reproduction of H.axyridis larvae.The results of the study will help provide theoretical support for further research on the relationship between key gene expression and energy metabolism of H.axyridis larvae in the future and provide technical guidance and reference for rational use of H.axyridis in biological control.
