越南槐种子萌发响应钙离子胁迫的差异表达蛋白分析

摘要 为分析钙离子胁迫下越南槐种子萌发的分子响应机制,以越南槐种子为材料,在0、50、100 mmol/L CaCl_(2)溶液处理下直至萌发,利用非标记定量蛋白质组学技术结合质谱鉴定和生物信息学分析越南槐种子萌发响应钙离子胁迫的差异表达蛋白。结果表明,50 mmol/L CaCl_(2)vs 0 mmol/L CaCl_(2)比较组中,表达上调的蛋白有43个,表达下调的蛋白有42个;100 mmol/L CaCl_(2)vs 0 mmol/L CaCl_(2)比较组中,表达上调的蛋白有44个,表达下调的蛋白有37个;100 mmol/L CaCl_(2)vs 50 mmol/L CaCl_(2)比较组中,表达上调的蛋白有32个,表达下调的蛋白有21个。差异表达蛋白主要参与信号转导,DNA复制和转录,蛋白质合成、加工和降解,糖与能量代谢,脂代谢,次生代谢,细胞骨架及细胞壁代谢,抗氧化及防御等过程。通过高通量蛋白质组学分析,初步揭示了参与越南槐种子萌发应答钙离子胁迫的关键蛋白质。 In order to analyze the molecular response mechanism of Sophora tonkinensis seed germination under calcium ion stress.The S.tonkinensis seeds were treated with 0,50 and 100 mmol/L CaCl_(2)solution until germination and the germinated seeds were collected in this study.Label-free quantitative proteomics technology,combined with mass spectrometry and bioinformatics analysis,was used to analyze the differential expression proteins of S.tonkinensis seed germination in response to calcium stress.The results indicated that 43 proteins were up-regulated and 42 proteins were down-regulated in 50 mmol/L CaCl_(2)vs 0 mmol/L CaCl_(2)comparison group;in the 100 mmol/L CaCl_(2)vs 0 mmol/L CaCl_(2)comparison group,44 proteins were up-regulated and 37 proteins were down-regulated;in the 100 mmol/L CaCl_(2)vs 50 mmol/L CaCl_(2)comparison group,32 proteins were up-regulated and 21 proteins were down-regulated.The differentially expressed proteins were mainly involved in signal transduction,DNA replication and transcription,protein synthesis,processing and degradation,carbohydrate and energy metabolism,lipid metabolism,secondary metabolism,cytoskeleton and cell wall metabolism,antioxidant and defense processes.Through high-throughput proteomic analysis,the key proteins involved in the seed germination response to calcium stress of S.tonkinensis were preliminarily revealed.
