lncRNA BC002811通过调控巨噬细胞表型转换对卵巢癌细胞生物学行为的作用机制

摘要 目的:探究lncRNA BC002811通过调控巨噬细胞表型转化对卵巢癌细胞生物学行为的作用机制。方法:将SKOV3细胞分为sh-NC组、sh-BC002811组和blank组,RT-PCR检测各组细胞中lncRNA BC002811表达和M0巨噬细胞中CD68表达。分别采用MTT法、Transwell法检测细胞增殖和侵袭能力。建立巨噬细胞和卵巢癌细胞的共培养体系,检测巨噬细胞表面CD86、CD206的表达,检测巨噬细胞上清液中炎症相关因子水平。将细胞分为mi-BC002811组、mi-NC组、SKOV3组,检测过表达lncRNA BC002811的SKOV3细胞对巨噬细胞向M2型分化、增殖及侵袭的影响。结果:与blank组相比,sh-BC002811组细胞中lncRNA BC002811表达显著降低(P<0.05)。sh-BC002811组细胞增殖及侵袭能力均明显低于blank组(P<0.05)。与blank组相比,sh-BC002811组M1型巨噬细胞表面标志物CD86阳性比例升高,IL-6、TNF-α水平显著升高,M2型巨噬细胞表面标志物CD206阳性比例降低,IL-10水平显著降低(P<0.05)。与SKOV3组相比,mi-BC002811组M1型巨噬细胞表面标志物CD86阳性比例减少,IL-6、TNF-α水平显著降低,M2型巨噬细胞表面标志物CD206阳性比例增加,IL-10水平显著升高(P<0.05)。与SKOV3组相比,mi-BC002811组细胞增殖、侵袭能力显著增加(P<0.05)。结论:沉默lncRNA BC002811可通过抑制巨噬细胞向M2型分化,进而抑制卵巢癌细胞增殖分化。 Objective:To explore the mechanism of lncRNA BC002811 on the biological behavior of ovarian cancer cells by regulating the phenotypic transformation of macrophages.Methods:SKOV3 cells were divided into sh-NC group,sh-BC002811 group and blank group.RT-PCR was used to detect the expression of lncRNA BC002811 in cells of each group and the expression of CD68 in M0 macrophages.Cell proliferation was detected by MTT assay,and cell invasion was detected by Transwell assay.The co-culture system of macrophages and ovarian cancer cells was established.The expressions of CD86 and CD206 on the surface of macrophages were detected,and the levels of inflammatory related factors in the supernatant of macrophages were detected.The cells were divided into mi-BC002811 group,mi-NC group and SKOV3 group,and the effect of SKOV3 cells overexpress lncRNA BC002811 on the differentiation,proliferation and invasion of macrophages into M2 type was detected.Results:Compared with blank group,the expression of lncRNA BC002811 in sh-BC002811 group was significantly decreased(P<0.05).Compared with blank group,cell proliferation and invasion ability of sh-BC002811 group were significantly decreased(P<0.05).Compared with blank group,the positive ratio of M1 macrophage surface marker CD86 in sh-BC002811 group was increased,the levels of IL-6 and TNF-αwere increased significantly,the positive ratio of M2 macrophage surface marker CD206 was decreased,and IL-10 level was significantly lower(P<0.05).Compared with SKOV3 group,the positive ratio of M1 macrophage surface marker CD86 in mi-BC002811 group decreased,the levels of IL-6 and TNF-αwere significantly decreased,the positive ratio of M2 macrophage surface marker CD206 was increased,and IL-10 level was significantly increased(P<0.05).Compared with SKOV3 group,the cell proliferation and invasion ability of mi-BC002811 group were significantly increased(P<0.05).Conclusion:Silencing lncRNA BC002811 can inhibit the proliferation and differentiation of ovarian cancer cells by inhibiting the differentiation of macrophages to M2 type.
