北方晚熟马铃薯病毒抗性鉴定及分子标记检测

Evaluation of Virus Resistance and Detection of Markers for Disease Resistance Genes in Late-maturing Potato Varieties from North China

摘要 病毒病等病害是中国马铃薯产业的重要限制因子。为了马铃薯病毒病等病害的防治,研究通过病毒接种鉴定了15个北方晚熟马铃薯品种(系)对马铃薯Y病毒(Potato virus Y,PVY)、马铃薯X病毒(Potato virus X,PVX)、马铃薯S病毒(Potato virus S,PVS)和马铃薯A病毒(Potato virus A,PVA)四种主要马铃薯病毒的抗性,同时检测了这些品种部分病害的相关分子标记。有10份试验材料至少对一种病毒表现出抗性,其中‘京张薯4号’‘冀张薯8号’‘冀张薯12号’‘2013-19-14’和‘2013-89-2’同时具有3种病毒抗性,表明京张薯和冀张薯系列马铃薯具有较好病毒综合抗性。标记检测结果显示,‘冀张薯8号’‘2013-38-32’和‘2013-89-2’的PVY抗性可能来源分别为Ry_(adg)、Ry_(chc)、Ry_(sto),而‘冀张薯12号’的PVX抗性可能来源于Rx1。根据标记与抗性表型的符合程度推测,上述马铃薯品种(系)的PVY抗性可能主要来源Ry_(sto),PVX抗性可能主要来源Rx1。分子标记还检测出6份材料含有马铃薯根结线虫抗性基因H1和4份材料晚疫病抗性基因R8。研究为这些抗性资源的进一步利用以及多种抗性基因的聚合育种奠定了基础。 Viral and other diseases are important limiting factors of the potato industry in China.For management of these diseases in potatoes,the resistances against potato virus Y(PVY),potato virus X(PVX),potato virus S(PVS),and potato virus A(PVA)were evaluated in 15 late-maturing potato varieties(lines)from Northern China by mechanical inoculation in this study,and the diseases related molecular markers were also detected.Ten potato varieties(lines)exhibited resistance to at least one of the tested viruses,among which'Jingzhangshu 4','Jizhangshu 8','Jizhangshu12','2013-19-14',and'2013-89-2'were resistant to three of four tested viruses,indicating that these potatoes expressed good comprehensive resistance to viruses.The marker detection results revealed that the PVY resistance in'Jizhangshu 8','2013-38-32',and'2013-89-2'were possibly derived from Ry_(adg),Ry_(chc),and Ry_(sto),respectively,while the PVX resistance in'Jizhangshu 12'was probably derived from Rx1.According to the degree of coincidence between markers and phenotypes,the PVY and PVX resistance in these potato varieties(lines)may be mainly from Ry_(sto),and Rx1,respectively.Molecular markers also detected potato root-knot nematode resistance gene H1 in six potato varieties(lines)and potato late blight resistance gene R8 in four potato varieties(lines).This study may lay a foundation for the further utilization of these resistance resources and the pyramid breeding of multiple resistance genes.
