基于网络药理学研究羌活-菊花药对治疗冠心病的作用机制

摘要 目的基于网络药理学初探羌活-菊花药对治疗冠心病的作用机制。方法运用中药系统药理学计数分析平台以及GeneCards、Uniprot等数据库筛选羌活、菊花的有效活性成分及靶标。运用Cytoscape软件将活性成分-靶标网络进行可视化,经STRING数据库构建蛋白互作网络,进行基因本体(GO)分析以及京都基因与基因组百科全书(KEGG)通路富集分析。结果获得羌活、菊花主要活性成分共74个,潜在靶点757个,冠心病相关靶点共3687个,包括HTR3A、NCOA2、KCNH2等关键靶点,β-谷甾醇、芹菜素等主要成分,KEGG富集分析主要涉及HIF-1、MAPK、FoxO等信号通路,GO富集分析生物过程主要包括对镉离子的反应、对活性氧的反应等,细胞成分包括薄膜筏子、膜微域等,分子功能富集倍数较大的有核受体活性、配体激活的转录因子活性等。结论本研究初步揭示了羌活-菊花药对治疗冠心病的主要活性成分、关键靶点及其作用机制。 Objective To explore the mechanism of action of Notopterygium incisum-Chrysanthemum morifolium in the treatment of coronary heart disease based on network pharmacology.Methods The active ingredients and targets of Notopterygium incisum and Chrysanthemum morifolium were screened using the systematic pharmacological count analysis platform of Chinese medicine and databases such as GeneCards and Uniprot.The active ingredient-target network was visualized using Cytoscape software.The protein inte-raction network was constructed by STRING database.The Gene Ontology(GO)enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis.Results A total of 74 main active ingredients of Notopterygium incisum and Chrysanthemum morifolium,757 potential targets and 3687 targets related to coronary heart disease were obtained,including key targets such as HTR3A,N-COA2,KCNH2,β-sitosterol,apigenin,and other major components,KEGG enrichment analysis mainly involved HIF-1,MAPK,FoxO,and other signaling pathways,GO enrichment analysis of biological processes mainly included the response to cadmium ions,reaction to reactive oxygen species,etc.,cellular components including membrane rafts,membrane microdomains,etc.,molecular functional enrichment multiples include nuclear receptor activity,ligand-activated transcription factor activity,etc.Conclusion This study initially reveals the main active components,key targets,and their mechanisms of action of the Notopterygium incisum-Chrysanthemum morifolium drug pair for the treatment of coronary heart disease,providing a scientific basis and research ideas for clinical use and treatment of coronary heart disease.
