活经汤对膝骨关节炎大鼠疼痛模型的治疗作用 被引量:1

Therapeutic effect of Huojing decoction(活经汤)on knee osteoarthritis in rats pain model

摘要 目的:探讨活经汤对单碘乙酸钠(MIA)诱导的膝骨关节炎(KOA)大鼠疼痛模型的治疗作用。方法:将24只健康成年雄性Sprague-Dawley大鼠随机分成4组,每组6只:对照组1(Saline组)、对照组2(Saline+活经汤组)、模型组(MIA组)和治疗组(MIA+活经汤组)。在大鼠膝关节腔内单次注射单碘乙酸(MIA)以建立大鼠KOA疼痛模型,对照组注射相同体积的生理盐水。在MIA注射后14 d进行连续的膝关节压力疼痛、膝关节伸展疼痛和机械痛行为学测试,以评估KOA疼痛模型的建立情况。对MIA诱导的KOA大鼠疼痛模型以2 ml浓度为50 mg/L活经汤悬液进行连续14 d的灌胃处理和疼痛行为学测试,并利用酶联免疫吸附实验测定各组大鼠背根神经节(DRG)中炎症细胞因子IL-1β、IL-6和TNF-α的表达水平。结果:与Saline组相比,在大鼠膝关节注射1 mg MIA能够在28 d内显著诱发大鼠膝关节产生连续的膝关节压力疼痛(4.2±0.8)次、膝关节伸展疼痛(4.0±0.9)次和机械痛敏行为(5.2±1.5)g( P<0.001)。以50 mg/L的活经汤悬液对KOA大鼠进行灌胃处理能够显著缓解由MIA诱导的大鼠KOA疼痛,使机械痛觉阈值显著增加(15.5±1.9)g,热痛阈值升高至(8.5±1.0)s,并使冷痛阈值恢复至(5.8±0.8)s( P<0.001)。同时,以50 mg/L的活经汤悬液对KOA大鼠进行灌胃处理能够显著降低DRG中炎症细胞因子IL-1β(41.3±11.1)pg/ml、IL-6(20.4±5.9)pg/ml和TNF-α(24.6±4.9)pg/ml的表达水平( P<0.05)。 结论:活经汤对单碘乙酸诱导的KOA疼痛大鼠模型具有良好的镇痛和抗炎作用。 Objective To explore the therapeutic effect of Huojing decoction(活经汤)on the rats pain model of knee osteoarthritis(KOA)induced by monosodium iodoacetate.Methods Twenty-four healthy adult male Sprague Dawley rats were randomly divided into 4 groups with 6 rats in each group:control group 1(saline group),control group 2(saline+Huojing decoction group),model group(MIA group)and treatment group(MIA+Huojing decoction group).A single injection of monosodium iodoacetate(MIA)was made in the knee joint cavity of rats to establish a model of KOA pain in rats.Continuous behavioral tests of knee pressure pain,knee extension pain and mechanical pain were performed 14 days after MIA injection to establish KOA pain model.The MIA-induced KOA rat pain model was administered with 2 ml of 50 mg/L Huojing decoction for 14 consecutive days of gastric administration.Finally,the enzyme-linked immunosorbent assay was used to determine the levels of inflammatory factors IL-1β,IL-6 and TNF-αin the dorsal root ganglia(DRG).Results Compared with saline group,injection of 1 mg MIA into rat knee could significantly induce continuous knee pressure pain[(4.2+0.8)times],knee extension pain[(4.0±0.9)times]and mechanical pain sensitivity behavior[(5.2±1.5)g]within 28 days(P<0.001).Intragastric administration of 50 mg/L Huojing decoction suspension to KOA rats could significantly alleviate the pain of knee arthritis induced by MIA,significantly increase the mechanical pain threshold by(15.5±1.9)g,the thermal pain threshold to(8.5±1.0)seconds,and restore the cold pain threshold to(5.8+0.8)seconds(P<0.001).Meanwhile,intragastric administration of 50 mg/L Huojing decoction suspension to KOA rats can significantly reduce the inflammatory cytokine IL-1β(41.3±11.1)pg/ml,IL-6(20.4±5.9)pg/ml and TNF-α(24.6±4.9)pg/ml(P<0.05).Conclusion Huojing decoction has effective analgesic and anti-inflammatory effects on MIA-induced rats KOA.
