摘要
目的:通过χ^(2)检验以及肠道菌群与不同中医体质的关系模型的构建,以证明肠道菌群与不同中医体质的相关性。方法:基于问卷调查对114名受试者进行体质分类,同时收集各受试者粪便,利用16S rRNA测序技术获得肠道菌群种水平层面相对丰度的定性数据,通过χ^(2)检验筛选出与不同中医体质关系密切的肠道差异菌群。基于基尼系数筛选出特征菌种并分别用朴素贝叶斯(NB)、K近邻(IBK)、支持向量机(SVM)、随机森林(RF)、反向传播神经网络(BPNN)5种算法进行肠道菌群与中医体质关系模型构建。结果:在P<0.05χ^(2)检验水平上,不同的肠道菌群与9种体质相关在一定程度上具有明显差异。基于基尼系数构建二者的关系模型,除气虚质外,其余体质都能构建关系模型,其受试者工作特征曲线下面积在0.7以上。结论:基于肠道菌群与不同中医体质构建的模型是合理可靠的,说明不同肠道菌群与9种中医体质具有一定的相关性。
Objective:To demonstrate the correlation between intestinal flora and different TCM constitutions by the chi-square test and the construction of a model of the relationship between intestinal flora and different TCM constitutions.Methods:Based on the questionnaire survey,114 subjects were classified by constitutions,the stool was collected from each subject to obtain qualitative data on the relative abundance of intestinal flora at the species level using 16S rRNA sequencing technology,and the intestinal differential flora that were closely related to different TCM constitutions were screened by chi-square test.Characteristic strains were screened based on the Gini coefficient and the relationship model between intestinal flora and TCM constitutions was constructed using five algorithms,namely NB,IBK,SVM,RF and BPNN,respectively.Results:At the level of chi-square test(P<0.05),there were significant differences in the relationship between different intestinal strains and nine constitutions to some extent.The relationship model between the two was constructed based on the Gini coefficient;the relationship model could be constructed for all constitutions except for the Qi deficiency type,whose area under the curve of the receiver operating characteristic was above 0.7.Conclusion:The model constructed based on intestinal flora and different TCM constitutions is reasonable and reliable,indicating that different intestinal flora have some correlation with nine TCM constitutions.
