新扶正除疫颗粒剂对感染腺病毒小鼠肺部炎性因子及 TLRs 信号途径的影响

Effects of New Fuzheng Chuyi Granules on pulmonary inflammatory factors and TLRs signal pathway in mice infected with adenovirus

摘要 目的探讨感染腺病毒小鼠使用新扶正除疫颗粒剂治疗后,肺内相关炎性因子及TLRs信号途径作用机制。方法雌性BALB/c小鼠120只,随机分为5组,新扶正除疫颗粒剂低、中、高剂量组(简称低、中、高剂量组)、腺病毒模型组、利巴韦林组。模型建立后,给药7天。最后1天摘得动物肺脏用荧光定量PCR法测白细胞介素-10(IL-10)、肿瘤坏死因子α(TNF-α)基因表达水平,Western blot、荧光定量PCR测定TLR9、MyD88和NF-κB蛋白与基因表达。结果比较模型组,低、中、高剂量组肺组织TNF-α基因表达显著下降(P<0.05,P<0.01),IL-10的基因表达显著增加(P<0.01),TLR9、MyD88和NF-κB观察指标蛋白与基因表达均下降(P<0.05,P<0.01),高剂量组下降最明显(P<0.01)。结论新扶正除疫颗粒剂保护被腺病毒侵袭的小鼠肺组织,可能的作用机制是调节TL R9、MyD88和NF-κB信号途径表达,以及调节TNF-α、IL-10的表达。 Objective To investigate the mechanism of lung related inflammatory factors and TLRs signal pathway in adenovirus infected mice treated with New Fuzheng Chuyi Granules.Methods R andomly divide 120 female BALB/c mice into 5 groups:low,medium,high dose groups of New Fuzheng Chuyi Granules,adenovirus group,ribavirin group.After the establishment of the infection model,the mice were administered continuously for 7 days,and the changes of mice were recorded.On the 7th day,the lungs of mice were taken,and the tumor necrosis factor-αwas measured by fluorescence quantitative PCR method,TLR9,MyD88 and NF-κB in lung were observed by fluorescence quantification PC R and Westernblot methods,to measure gene and protein expression level.Results Compared with the model group,TNF-αmRNA of every different dose groups expression declined(P<0.05,P<0.01),IL-10 mRNA elevated(P<0.01).The observation index of TLRs signaling pathway decreased(P<0.05,P<0.01),and the high-dose group the was most obvious(P<0.01).Conclusion New Fuzheng Chuyi Granules protect the lung tissue of mice invaded by adenovirus,and the possible mechanism is to regulated the expression of TLR9,MyD88 and NF-κB signal pathway and TNF-α,IL-10 expression.
