摘要
目的探讨氧化应激因子和铁死亡标志物在子痫前期孕妇中的表达情况及其临床意义。方法选取30例子痫前期孕妇作为子痫前期组,30例正常孕妇作为对照组。检测两组孕妇外周血中氧化应激因子超氧化物歧化酶(SOD)、丙二醛和谷胱甘肽表达水平。观察两组孕妇胎盘组织的病理学变化,并检测胎盘组织中铁死亡标志物谷胱甘肽过氧化物酶4(GPX4)和环氧合酶2(COX2)的蛋白和mRNA表达水平。结果子痫前期组孕妇外周血中SOD、谷胱甘肽表达水平低于对照组,丙二醛表达水平高于对照组(均P<0.05)。对照组孕妇胎盘组织的细胞轮廓清晰且排列整齐,绒毛组织几乎无破损;子痫前期组孕妇的胎盘组织出现细胞溶解和血管水肿破裂的现象。子痫前期组孕妇的胎盘组织中GPX4蛋白及mRNA表达水平均低于对照组,而COX2蛋白及mRNA表达水平均高于对照组(均P<0.05)。结论子痫前期孕妇外周血中的氧化应激因子和胎盘组织中的铁死亡标志物表达异常,氧化应激反应及铁死亡可能参与子痫前期的发病过程。
Objective To investigate the expressions of oxidative stress factors and ferroptosis markers in pregnant women with preeclampsia and its clinical significance.Methods A total of 30 pregnant women with preeclampsia were selected as preeclampsia group,and 30 normal pregnant women as control group.The expressions of oxidative stress factors in terms of superoxide dismutase(SOD),malondialdehyde,and glutathione in peripheral blood were detected in pregnant women of the two groups.Pathological change of placental tissues in pregnant women of the two groups was observed,and protein and mRNA expressions of ferroptosis markers with respect to glutathione peroxidase 4(GPX4)and cyclooxygenase 2(COX2)in placental tissues were detected in pregnant women of the two groups.Results Pregnant women in the preeclampsia group exhibited lower expressions of SOD and glutathione,and a higher expression of malondialdehyde in peripheral blood as compared with the control group(all P<0.05).Cells in the placental tissues of pregnant women in the control group presented as clearly defined outline,and were neatly arranged,as well as presented as almost undamaged villous tissues,while the placental tissues of pregnant women in the preeclampsia group revealed cytolysis,and vascular edema rupture.Pregnant women in the preeclampsia group interpreted lower protein and mRNA expressions of GPX4 in placental tissues,whereas higher protein and mRNA expressions of COX2 in placental tissues as compared with the control group(all P<0.05).Conclusion The expressions of oxidative stress factors in peripheral blood and ferroptosis markers in placental tissues of pregnant women with preeclampsia are abnormal,and oxidative stress response and ferroptosis may be involved in the pathogenetic process of preeclampsia.
