角果碱蓬响应干旱胁迫的转录组分析 被引量:3

Transcriptomic analysis of Suaeda corniculata in response to drought stress

摘要 角果碱蓬(Suaeda corniculata)是一种特殊逆境适生植物,目前其应答干旱胁迫的调节机制仍不清楚。本研究针对角果碱蓬的耐旱特性,通过模拟干旱胁迫,测定了角果碱蓬幼苗光合参数与相对含水量,系统分析响应干旱胁迫的基因表达谱,以期挖掘角果碱蓬响应干旱胁迫的关键调节基因。结果表明,角果碱蓬对干旱胁迫有一定的耐受能力,尽管干旱处理降低了光合作用效率和相对含水量,但胁迫状态解除后能够很快恢复到正常水平。转录组分析共鉴定到720个基因在干旱胁迫下发生差异表达,其中上调表达基因426个,下调表达基因294个,它们主要富集在细胞代谢、遗传信息处理和环境适应等生物学过程。对差异表达基因进一步筛选,获得了转录因子基因(c139038.graph_c2和c140552.graph_c1)以及ABA激素信号基因(c125016.graph_c0和c147554.graph_c0)等潜在的干旱耐受相关调节因子。研究结果为角果碱蓬干旱适生生理机制解析提供了新的证据,并为其抗旱性研究提供了分子基础。 Suaeda corniculata is a special adversity plant,and its regulatory mechanism in response to drought stress is still unclear.Based on its drought-tolerant characteristics,in this study,the photosynthetic parameters and relative water content of S.corniculata were measured,and the gene expression profiles were systematically analyzed to explore the key regulatory genes of S.corniculata in response to drought stress.The results showed that S.corniculate has a certain tolerance to drought stress,although the photosynthesis efficiency and relative water content were reduced by the drought treatment,they could quickly recover to the normal level after re-watering.A total of 720 differentially expressed genes were identified by transcriptomic sequencing in S.corniculata under drought stress(including 426 up-regulated and 294 down-regulated genes),and these genes mainly enriched in several biological processes such as cellular metabolism,genetic information processing,and environmental adaptation.We further analyzed the differentially expressed genes,and potential drought tolerance-related regulators such as transcription factor genes(c139038.graph_c2 and c140552.graph_c1)and ABA-induced signal gene(c125016.graph_c0 and c147554.graph_co)were obtained.This study provides new evidence for understanding the physiological mechanism of S.corniculata adapt to drought and a molecular basis for the study on drought resistance.
