摘要
变孔异担子菌(Heterobasidion irregulare)是一种侵染性强、危害大的针叶树病原菌。为建立变孔异担子菌的KASP检测方法,本研究根据变孔异担子菌GAPDH基因的SNP位点,筛选出变孔异担子菌KASP分子标记,并检测了该标记的特异性、灵敏度。结果表明,引物Hirr-FAM-F1、Hirr-HEX-F2、Hirr-R可以准确鉴定变孔异担子菌,检测灵敏度可达0.5 ng/μL。
Heterobasidion irregulare is a fungus with strong infection and great hazard on coniferous wood.In order to establish a KASP detection method for H.irregulare,this study screened out a molecular marker of H.irregulare based on the SNP site of the GAPDH gene,and the specificity and sensitivity were tested.The results showed that Hirr-FAM-F1、Hirr-HEX-F2、Hirr-R could accurately identify H.irregulare and the detection sensitivity was up to 0.5 ng/μL.
