IL-33及其受体ST2在前列腺癌组织中的表达及意义

摘要 目的 探讨白细胞介素33(interleukin 33,IL-33)及其受体生长刺激表达基因2蛋白(growth stimulation expressed gene 2 protein, ST2)在正常前列腺组织(normal prostate, NP)、良性前列腺增生组织(benign prostatic hyperplasia, BPH)以及前列腺癌(prostate cancer, PCa)组织中的表达差异.方法 收集癌旁正常前列腺组织4例,良性前列腺增生组织53例,前列腺癌组织42例;通过免疫组织化学染色技术检测各个组织中IL-33及ST2的阳性表达率,并分析二者之间关系,同时判定IL-33和ST2与前列腺癌恶性度之间的关系;通过免疫组织化学染色法检测CD68^(+)巨噬细胞在不同前列腺组织的浸润差异,并分析IL-33与巨噬细胞浸润的关系.结果 与NP相比,PCa组织中IL-33和ST2表达明显降低,且随前列腺癌Gleason分级增加,其表达降低(P<0.05);与BPH相比,PCa组织中IL-33和ST2表达明显降低(P<0.05);IL-33与ST2在BPH、PCa中的表达均呈正相关关系;IL-33与CD68^(+)巨噬细胞在BPH、PCa中的表达均呈正相关关系.结论 BPH和PCa中的IL-33可能是由巨噬细胞分泌的,IL-33可能具有抑制前列腺癌发生发展的作用. Objective To investigate the expression differences of interleukin 33(IL-33)and its receptor growth stimulation expressed gene 2 protein(ST2)in normal prostate(NP),benign prostatic hyperplasia(BPH)and prostate cancer(PCa)tissues and explore the clinical significance.Method 4 paracancerous normal prostate tissues,53 benign prostatic hyperplasia tissues and 42 prostate cancer tissues were collected and immun-ohistochemical staining technique was applied to detect the expression of IL-33 and ST2,and analyze the relationship between of them.The malignancy of prostate cancer between IL-33 and ST2 was analyzed simul-taneously.CD68^(+)macrophages were stained in different prostatic tissues by using immunohistochemical staining method.The correlation between IL-33 and macrophages infiltration was analyzed.Results Compared with NP,IL-33 and ST2 expression were significantly lower in PCa tissues and had significantly statistical differences(P<0.05).The expression of IL-33 and ST2 decreased with Gleason grade increasing.Compared with BPH,IL-33 and ST2 expression were significantly lower in PCa tissues and had significantly statistical differences(P<0.05),IL-33 and ST2 in BPH and PCa are positively correlated;IL-33 and CD68^(+)macrophages in BPH and PCa are positively correlated respectively.Conclusion IL-33 may be secreted from CD68^(+)macrophages and have an inhi-bitory effect on prostate cancer occurrence and development.
