摘要
目的:探讨大鼠多囊卵巢综合征(PCOS)模型表皮-钙黏蛋白(E-cadherin)、增殖细胞核抗原(PCNA)表达情况及意义。方法:6周龄SD清洁级雌性大鼠12只,随机分为来曲唑处理组((印)n(正)=6)和正常对照组((印)n(正)=6),来曲唑处理组用来曲唑1 mg/kg灌胃造模,正常对照组给予同等剂量溶剂灌胃,连续处理21天后,麻醉剂量处死,HE染色观察卵巢形态,免疫组织化学染色方法检测E-cadherin、PCNA蛋白定位,蛋白印迹检测E-cadherin、PCNA蛋白表达。结果:正常对照组卵巢视野中可见各阶段发育卵泡,来曲唑处理组卵巢仅有多个大小不等的空卵泡生长,内颗粒细胞层数较对照组明显减少;E-cadherin、PCNA蛋白均定位在细胞质和细胞核;蛋白印迹检测发现,PCNA蛋白来曲唑处理组表达明显低于正常对照组,E-cadherin蛋白来曲唑处理组表达高于正常对照组,且差异均有统计学意义((印)P(正)<0.05)。结论:来曲唑诱导大鼠PCOS模型卵巢中,PCNA蛋白表达降低,E-cadherin蛋白表达增加,可能使卵巢颗粒细胞增殖能力下降。
Objective:To investigate the expression and significance of E-cadherin and PCNA in rat PCOS model.Methods:Twelve 6-week-old SD clean grade female rats were randomly divided into two groups:letrozole treatment group(n=6)and normal control group(n=6).Letrozole treatment group was given 1 mg/kg of letrozole for intragastric modeling.while normal control group was given the same dose of solvent for intragastric modeli ng.After 21 days of continuous treatment,the rats were sacrificed by anesthetized dose,and the ovarian morphology was observed by HE staining.Immunohistochemical staining was used to detect the localization of E-cadherin and PCNA proteins,and Western blotting was used to detect the expression of E-cadherin and PCNA proteins.Results:!n the control group,ovarian follicles at different stages of development were observed in the field of vision.In the letrozole treatment group,only a number of empty follicles of different sizes grew,and the number of inner granular cell layers was significantly reduced compared with that in the control group.E-cadherin and PCNA proteins were localized in cytoplasm and nucleus.Western blotting showed that the expression of PCNA protein in letrozole treatment group was significantly lower than that in the control group,and the expression of E-cadherin protein in letrozole treatment group was higher than that in the control group,with statistically significant differ-ences(P<0.05).Conclusions:In the ovaries of PCOS model induced by letrozole Jhe expression of PCNA protein decreased and the expression of E-cadherin protein increased,which resulted in the decreased proliferation of ovarian granulosa cells.
