萝卜种质亲缘关系的分子标记分析

Molecular Marker Analysis on Genetic Relationship of Radish Germplasms

本研究以118个分子标记在108份萝卜种质中进行筛选,得到56个带型清晰、多态性好的标记用于亲缘关系分析。用这56个标记共扩增出130条多态性条带,每个标记2~5条,平均2.3条;观测等位基因数的平均值为2.3214,有效等位基因数的平均值为1.8643,Nei基因多样性指数的平均值为0.4573,Shannon多态性指数的平均值为0.6766,PIC值介于0.29~0.55之间。UPGMA聚类和PCA分析结果较为一致,在遗传相似系数为0.61处可将108份种质分为4大类,黑皮白肉萝卜、樱桃萝卜、心里美类型萝卜种质各聚为一类,其余种质聚为一类,表明萝卜种质亲缘关系与肉质根皮色、肉质色、大小的联系较密切。

In this study,118 molecular markers were used and 56 ones with clear bands and polymorphism were selected for genetic relationship analysis of 108 radish germplasms.A total of 130 polymorphic bands were amplified using these 56 markers,2 to 5 bands were amplified for each marker,with an average of 2.3 bands;the mean value of observed alleles was 2.3214 and the mean value of effective alleles was 1.8643;the mean value of Nei’s expected heterozygosity was 0.4573 and the mean value of Shannon’s information index was 0.6766;the PIC value of the 56 markers ranged from 0.29 to 0.55.Consistent results were obtained from UPGMA clustering and PCA analysis,the 108 germplasms were divided into four groups with clustering threshold of 0.61;the black skin and white meat radish,cherry radish and Xinlimei radish germplasms each clustered into one category,while the rest of the germplasms clustered into one category,indicating that genetic relationship was closely related to skin color,flesh color and size of taproot.
