摘要
研究七氟醚调控丝裂原活化蛋白激酶(MAPK)信号通路对不同孕期大鼠子代认知功能的影响。将80只大鼠随机分为4个组别:对照组、孕早期(孕6 d)组、孕中期(孕12 d)组和孕晚期(孕18 d)组(N=20)。吸入七氟醚(3%),子代出生7 d后进行水迷宫、回避和避暗实验。通过TUNEL染色和蛋白免疫印迹检测海马神经元凋亡和P38 MAPK通路水平。实验结果显示:孕早期组的子代学习记忆功能显著低于对照组,回避时间和错误次数、海马神经元凋亡情况、P38 MAPK蛋白磷酸化水平显著高于对照组(P<0.05);孕中期组的子代学习记忆功能显著低于孕早期组,回避时间和错误次数、海马神经元凋亡情况、P38 MAPK蛋白磷酸化水平显著高于孕早期组(P<0.05);孕晚期和对照组各项指标比较差异无统计学意义(P<0.05)。研究表明,孕早期和孕中期吸入七氟醚会激活子代P38 MAPK通路诱导海马神经元凋亡,对子代认知功能产生负面影响。
The study purpose was to explore the effect of sevoflurane-regulated MAPK signaling pathway on the cognitive function of the offspring of rats in different pregnancy periods.Eighty rats were randomly divided into 4 groups:the control group,the first trimester(6 days of pregnancy)group,the second trimester(12 days of pregnancy)and the third trimester(18 days of pregnancy)group(N=20),inhalation of sevoflurane(3%).The water maze,avoidance and dark avoidance experiments were taken at 7 days after the offspring'birth.The apoptosis of hippocampal neurons and the level of P38 MAPK pathway were detected by TUNEL staining and Western blot.It was showed that the learning and memory function of the offspring from the early pregnancy group was significantly lower than that of the control group,the avoidance time,the number of errors,the apoptosis of hippocampal neurons and the phosphorylation level of P38 MAPK protein were significantly higher than those of the control group(P<0.05).The learning and memory function of the offspring from the second trimester group was significantly lower than that of the early trimester group,the avoidance time,the number of errors,hippocampal neuron apoptosis,and P38 MAPK protein phosphorylation level were significantly higher than those of the early pregnancy group(P<0.05).There was no statistically significant difference in indicators between the third trimester of pregnancy and the control group(P<0.05).Inhalation of sevoflurane in the first and second trimesters will activate the offspring's P38 MAPK pathway to induce hippocampal neuronal apoptosis,which has a negative impact on offspring's cognitive function.
