异紫堇碱对肺癌A549细胞增殖、迁移和侵袭的作用

摘要 目的探讨异紫堇碱对肺癌A549细胞增殖、迁移和侵袭的影响及其分子机制。方法将A549细胞分为对照组,异紫堇碱低、中、高剂量组,miR-NC组,miR-1296-5p组,anti-miR-NC组,anti-miR-1296-5p组,异紫堇碱+anti-miR-NC组,异紫堇碱+anti-miR-1296-5p组。MTT法检测细胞增殖抑制率,Transwell实验检测细胞迁移和侵袭能力,RT-qPCR法检测miR-1296-5p和花斑抑制因子同源物1(SUV39H1)mRNA表达,Western blot法检测相关蛋白表达,双荧光素酶报告实验验证miR-1296-5p和SUV39H1的靶向关系。结果不同剂量异紫堇碱处理A549细胞后,细胞增殖抑制率升高,迁移、侵袭细胞数降低,miR-1296-5p表达升高,SUV39H1 mRNA和蛋白表达降低,并呈剂量依赖性(P<0.05)。miR-1296-5p靶向调控SUV39H1,过表达miR-1296-5p可抑制肺癌细胞增殖、迁移和侵袭。结论异紫堇碱可能通过调控miR-1296-5p/SUV39H1抑制肺癌细胞增殖、迁移和侵袭。 AIM To investigate the effects and molecular mechanism of isocorydine on the proliferation,migration and invasion of lung cancer cells.METHODS A549 cells were divided into the control group,low-dose,medium-dose and high-dose isocorydine groups,miR-NC group,miR-1296-5p group,anti-miR-NC group,anti-miR-1296-5p group,isocorydine+anti-miR-NC group,and isocorydine+anti-miR-1296-5p group.The A549 cells had their proliferation inhibition rate detected by MTT assay;their migration and invasion abilities detected by Transwell assay;their mRNA expressions of miR-1296-5p and stain inhibitor homologue 1(SUV39H 1)detected by RT-qPCR;their expressions of related proteins detected by Western blot;and their targeting relationship between miR-1296-5p and SUV39H1 verified by a double luciferase reporter assay.RESULTS A549 cells treated with different doses of isocorydine shared increased proliferation inhibition rate,decreased numbers of migrating and invasive cells,increased miR-1296-5p expression,decreased expression of SUV39H1 mRNA and protein(P<0.05)in a dose-dependent manner.Being an SUV39H1 regulator,miR-1296-5p inhibited the proliferation,migration and invasion of lung cancer cells if at the overexpression level.CONCLUSION Isocorydine may inhibit the proliferation,migration and invasion of lung cancer cells by regulating miR-1296-5p/SUV39H1.
