摘要
目的探讨JQ-1与小豆蔻明(CAR)联用对乳腺癌细胞MCF-7增殖和细胞凋亡的影响。方法将MCF-7细胞实验分为空白组(不给药)、对照组(10.00μmol·L^(-1)CAR)、实验组(25.00μmol·L^(-1)JQ-1)及联合组(10.00μmol·L^(-1)CAR+25.00μmol·L^(-1)JQ-1)。4组细胞均培养48 h。用CCK-8法检测细胞的增殖能力,用Annexin-V FITC/PI双染法检测细胞的凋亡情况,用蛋白质印迹法检测磷酸化磷脂酰肌醇3激酶(p-PI3K)、磷酸化蛋白激酶B(p-AKT)和活化后胱天蛋白酶7(Cleaved Caspase7)的表达水平。结果联合组、实验组、对照组和空白组的增殖率分别为(47.79±11.27)%、(66.75±26.56)%、(74.57±13.59)%和(100.00±0)%,细胞晚期凋亡率分别为(24.17±1.27)%、(7.07±0.80)%、(14.70±3.12)%和(4.98±2.98)%,p-PI3K蛋白相对表达水平分别为0.87±0.02、1.11±0.06、1.09±0.03和1.00±0.00,p-AKT蛋白相对表达水平分别为0.31±0.05、0.96±0.06、0.96±0.16和1.00±0.00,Cleaved Caspase7蛋白相对表达水平分别为2.70±0.04、1.51±0.03、1.28±0.01和1.00±0.00。联合组的上述指标与空白组、对照组、实验组比较,差异均有统计学意义(P<0.05,P<0.01)。结论JQ-1与CAR联用对乳腺癌细胞MCF-7具有增强抑制细胞增殖、促进细胞凋亡的作用,其作用机制可能与调控PI3K/AKT通路相关。
Objective To explore the effect of JQ-1 combined with cardamomin(CAR)on the proliferation and apoptosis of breast cancer MCF-7 cells.Methods MCF-7 cells were divided into blank group(without drug),control group(10.00μmol·L^(-1)CAR),experimental group(25μmol·L^(-1)JQ-1)and combined group(10.00μmol·L^(-1)CAR+25.00μmol·L^(-1)JQ-1).Four groups were cultured for 48 hours.CCK-8 was adopted to detect cell proliferation.The cell apoptosis was analyzed by Annexin-V FITC/PI.Western blot was used to detect the expression levels of phosohp-phosphoinositide 3-kinase(p-PI3K),phospho-protein kinase B(p-AKT)and Cleaved Caspase7.Results The proliferation rates of blank,control,experimental and combined groups were(47.79±11.27)%,(66.75±26.56)%,(74.57±13.59)%and(100.00±0)%;the later apoptosis rates were(24.17±1.27)%,(7.07±0.80)%,(14.70±3.12)%and(4.98±2.98)%;the relative expression levels of p-PI3K protein were 0.87±0.02,1.11±0.06,1.09±0.03 and 1.00±0.00;the relative expression levels of p-AKT protein were 0.31±0.05,0.96±0.06,0.96±0.16 and 1.00±0.00;the relative expression levels of Cleaved Caspase7 protein were2.70±0.04,1.51±0.03,1.28±0.01 and 1.00±0.00,respectively.The differences were statistically significant between the combined group and the blank,control,experimental groups(P<0.05,P<0.01).Conclusion JQ-1combined with CAR has a synergistic effect on breast cancer cell MCF-7,which can enhance the inhibition of cell proliferation and promote cell apoptosis.The mechanism may be related to the regulation of PI3K/AKT signal pathways.
