基于铜死亡相关基因的慢加急性肝衰竭的机制研究

摘要 目的 探究铜死亡相关基因在慢加急性肝衰竭(Acute-on-chronic liver failure, ACLF)发生发展中的作用,从而进一步研究ACLF与铜死亡相关的发病机制。方法 从GEO(Gene Expression Omnibus, GEO)数据库中下载ACLF相关数据。利用R软件筛选GEO数据库中ACLF与慢性肝病对比的差异表达基因(Differentially expressed genes, DEGs)和铜死亡相关基因的取交集,对共同差异基因通过GSEA(Gene Set Enrichment Analysis,基因集富集分析)分别进行单基因GO/KEGG富集分析,对相关基因通路富集结果结合铜死亡代谢图绘制铜死亡相关ACLF机制图。结果 共筛选出3个关于铜死亡相关的ACLF差异表达基因(P<0.05),进一步分析得出与铜死亡相关ACLF发生发展机制。结论 通过生物信息学分析,筛选出可能参与ACLF发病机制的铜死亡相关基因和信号通路,以期为ACLF的发病机制的研究提供理论基础以及对ACLF的发生发展和治疗提供新的靶点和策略。 Objective To investigate the role of copper death-related genes in the occurrence and development of acute-on-chronic liver failure(ACLF),so as to further study the pathogenesis of ACLF and copper death.Methods The ACLF-related data was downloaded from the GEO(Gene Expression Omnibus)database.R software was used to screen the intersection of differentially expressed genes(DEGs)and copper death-related genes compared with ACLF and chronic liver disease in the GEO database,and single-gene GO/KEGG enrichment analysis was performed through GSEA(Gene Set Enrichment Analysis).The enrichment results of related gene pathways combined with the metabolic map of copper death were used to map the mechanism of ACLF associated with copper death.Results A total of three differentially expressed genes of ACLF associated with copper death(P<0.05)were screened,and the mechanism of ACLF associated with copper death was further analyzed.Conclusion Through bioinformatics analysis,the genes and signaling pathways related to copper death may be involved in the pathogenesis of ACLF are screened,so as to provide a theoretical basis for the study of the pathogenesis of ACLF and new targets for the occurrence,development and treatment of ACLF.
