三角褐指藻胞内多糖提取条件优化及其抗氧化和抗肿瘤活性研究 被引量:2

Optimization of Extraction Conditions and Antioxidant, Antineoplastic Activity of Intracellular Polysaccharides from Phaeodactylum Tricornutum

摘要 为研究三角褐指藻胞内多糖的提取工艺及其生物活性,以多糖提取率作为指标,结合单因素和响应面法优化三角褐指藻多糖提取条件,对提取的多糖在体外进行抗氧化及抗肿瘤活性试验。结果表明,三角褐指藻胞内多糖最优提取条件为Na OH质量分数2.5%、料液比1:25 g/mL、破壁功率250 W、破壁时间5 min、水浴温度60℃、水浴时间1.5 h,在此条件下,胞内多糖的提取率为(2.315±0.01)%。抗氧化试验结果显示,三角褐指藻胞内多糖对DPPH自由基的清除率最大达到(95.09±1.05)%,对羟基自由基清除率最大达到(73.67±1.69)%,对超氧阴离子抑制率达到(70.34±2.75)%。三角褐指藻多糖抑制癌细胞增殖试验结果表明,该多糖抑制肝癌细胞HePG2、乳腺癌细胞MCF-9、结肠癌细胞HT-29增殖均有显著效果,最大抑制率分别为(62.77±0.99)%、(54.79±1.21)%、(45.26±1.79)%。以上结果显示,三角褐指藻胞内多糖具有良好的体外抗氧化及抗肿瘤活性。 In order to study the extraction process and biological activity of intracellular polysaccharide from Phaeodactylum tricornutum,the extraction conditions were optimized by single factor and response surface methodology with the extraction rate of polysaccharide as index.The antioxidant and antitumor activities of polysaccharide were tested in vitro.The results showed that the optimal extraction conditions of intracellular polysaccharide from Phaeodactylum tricornutum were as follows:NaOH mass fraction 2.5%,solid-liquid ratio 1:25 g/mL,wall-breaking power 250 W,wall-breaking time 5 min,water bath temperature 60 C,water bath time 1.5 h.Under these conditions,the extraction rate of intracellular polysaccharide was(2.315±0.01)%.The results of antioxidant experiments showed that the maximum scavenging rate of DPPH free radical was(95.09±1.05)%,the maximum scavenging rate of hydroxyl radical was(73.67±1.69)%,and the inhibition rate of superoxide anion was(70.34±2.75)%.The results of cancer cell proliferation inhibition test showed that polysaccharide had significant inhibitory effect on the proliferation of liver cancer cell HePG2,breast cancer cell MCF-9 and colon cancer cell HT-29,and the maximum inhibition rates were(62.77±0.99)%,(54.79±1.21)%and(45.26±1.79)%,respectively.The above results showed that the intracellular polysaccharide of Phaeodactylum tricornutum had good antioxidant an antitumor activities in vitro.
