SKA3在食管鳞状细胞癌中表达及其与临床病理参数的关系

摘要 目的探讨纺锤体与着丝粒相关蛋白3(SKA3)在食管鳞状细胞癌(ESCC)中的表达意义及其与临床病理参数的相关性。方法采用免疫组织化学染色法检测SKA3蛋白在75例食管鳞状细胞癌组织和20例正常食管组织中的表达,并分析其与临床病理特征的关系。应用癌症基因组图谱和基因芯片公共数据库分析SKA3 mRNA在203例食管鳞状细胞癌和102例正常对照中的表达。GEPIA在线工具分析SKA3与细胞周期调控基因的关系。Linkedomics及DAVID在线网站数据评价SKA3基因的共表达基因及其富集通路。结果在食管鳞状细胞癌组织中,SKA3 mRNA和蛋白的表达水平均高于正常食管上皮组织(P<0.05)。SKA3高表达与食管鳞状细胞癌患者肿瘤分化程度、临床分期及肿瘤神经侵犯密切相关(P<0.05)。SKA3与细胞周期调控基因CCND1、CDK1及CDK4的表达水平呈正相关(P<0.001)。结论在mRNA和蛋白水平上,SKA3在食管鳞状细胞癌中均高表达。SKA3高表达可能与食管鳞状细胞癌发生发展密切相关,其可能通过调控细胞周期进程而促进食管鳞状细胞癌的进展。 Objective To explore the significance of spindle and kinetochore associated protein 3(SKA3)expression in esophageal squamous cell carcinoma(ESCC)and its correlation with clinicopathologic parameters.Methods Immunohistochemical staining was used to detect the expression of SKA3 protein in 75 ESCC tissues and 20 normal esophageal tissues,and the relationship between SKA3 protein expression level and clinicopathological parameters was also analyzed.The Cancer Genome Atlas(TCGA)and Gene Expression Omnibus(GEO)were used to examine SKA3 mRNA expressions in ESCC(n=203)and normal controls(n=103).Moreover,GEPIA online tool was used to analyze the relationship between SKA3 and cell cycle-regulation genes.In addition,Linkedomics and DAVID online website data were used to evaluate the co-expression gene and enrichment pathway of SKA3 gene.Results Both the mRNA and protein levels of SKA3 in the ESCC tissues were all higher than those in the normal control esophageal tissues(P<0.05).The high expression of SKA3 was closely related to the degree of tumor differentiation,clinical stage and tumor nerve invasion in ESCC patients(P<0.05).And SKA3 was positively correlated with the expression levels of cell cycle-regulation genes including CCND1,CDK1 and CDK4(P<0.001).Conclusion SKA3 is highly expressed in ESCC at both mRNA and protein levels,which may be significantly correlated with the occurrence and development of ESCC.It may promote the progression of ESCC by regulating cell cycle progression.
