十枣汤UPLC指纹图谱化学计量学研究 被引量:2

摘要 目的 建立十枣汤指纹图谱研究方法,结合化学计量法对不同产地药材制成的十枣汤质量进行比较,完善十枣汤质量控制方法。方法 采用Waters T3 C18色谱柱(2.1 mm×100 mm,1.7μm),以乙腈-0.1%甲酸水溶液为流动相梯度洗脱,柱温40℃,流速0.4 mL/min,检测波长254 nm,采用《中药色谱指纹图谱相似度评价系统》(2012版)建立不同产地药材制成的13批十枣汤指纹图谱,利用相似度评价、主成分分析、聚类分析及偏最小二乘-判别分析对十枣汤样品进行质量评价。结果 建立了十枣汤UPLC指纹图谱,共确定31个共有峰,指认11个成分。13批十枣汤样品相似度良好,化学计量学方法可将其分为两类,十枣汤样品整体质量较好。结论 本研究建立的指纹图谱研究方法简便可靠,重复性良好,指纹图谱结合化学计量分析可明确不同产地药材制成的十枣汤之间的差异,为十枣汤的内在质量评价提供参考。 Objective To establish a method for studying the fingerprints of Shizao Decoction;To compare the quality of Shizao Decoction made from different producing areas of medicinal materials by chemometrics;To optimize the quality control method for Shizao Decoction.Methods Waters T3 C18 column(2.1 mm × 100 mm,1.7 μm) was used with acetonitrile-0.1% formic acid aqueous solution as mobile phase,gradient elution;flow rate was 0.4 mL/min;detection wavelength was 254 nm;column temperature was 40 ℃.13 batches of Shizao Decoction fingerprints made from different origins of medicinal materials were established by using Similarity Evaluation System of Chromatographic Fingerprint of TCM(2012 Edition).The quality of Shizao Decoction was evaluated by similarity evaluation,cluster analysis,principal component analysis and partial least square discriminant analysis.Results The UPLC fingerprints of Shizao Decoction was established.31 common peaks were identified and 11components were identified.The similarity of the samples was good.13 batches of Shizao Decoction were divided into two categories by chemometrics,and the overall quality of the samples was good.Conclusion The fingerprint research method established in the study is simple,reliable and reproducible.Through the method of fingerprint combined with chemometrics analysis,the differences between Shizao Decoction made from different origins of medicinal materials are identified,which provides a reference for the internal quality evaluation of Shizao Decoction.
