电针对大鼠肝缺血再灌注损伤保护作用的研究

Research on the protective effect of electroacupuncture on hepatic ischemia-reperfusion injury in rats

目的观察电针预刺激足三里和阳陵泉穴对大鼠肝脏缺血再灌注损伤的保护作用。方法将24只Sprague-Dawley(SD)雄性大鼠随机分为4组,模型组、非穴位组、穴位组和假手术组,每组6只。假手术组大鼠仅开腹腔暴露90 min后即关腹,模型组大鼠进行缺血90 min后再灌注处理,穴位组和非穴位组大鼠用电针预刺激30 min后再进行缺血90 min后再灌注处理。分别在再灌注4 h和24 h后,取大鼠肝脏组织进行苏木精-伊红(hematoxylin-eosin,HE)染色和组织髓过氧化物酶检测,比较血清丙氨酸转氨酶、天冬氨酸转氨酶、白介素-1β、白介素-6、白介素-10和肿瘤坏死因子-α的水平。结果再灌注4 h和24 h后,穴位组大鼠血清丙氨酸转氨酶、天冬氨酸转氨酶、白介素-1β、白介素-6和肿瘤坏死因子-α水平以及肝组织髓过氧化物酶表达均低于模型组(P<0.05),非穴位组上述指标与模型组比较差异无统计学意义(P>0.05)。结论电针预刺激足三里和阳陵泉穴对肝缺血再灌注损伤有一定的保护作用,可在提高肝功能、抑制炎症因子释放和减少中性粒细胞浸润方面发挥作用。

Objective To observe the protective effect of electroacupuncture at Zusanli(ST36)and Yanglingquan(GB34)on the hepatic ischemia-reperfusion(IR)injury in rats.Method Twenty-four Sprague-Dawley(SD)rats were randomly divided into four groups,including a model group,a non-point group,a point group,and a sham operation group,with 6 rats in each group.Rats in the sham operation group only received 90 min laparotomy without any other operation;rats in the model group received reperfusion after 90 min ischemia;rats in the point and non-point groups received 30 min electroacupuncture intervention before the 90 min ischemia and the following reperfusion.At 4 h and 24 h after the reperfusion,liver tissues of the rats underwent hematoxylin-eosin(HE)staining and myeloperoxidase(MPO)detection;the serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-10(IL-10),and tumor necrosis factor-α(TNF-α)were compared.Result At 4 h and 24 h after the reperfusion,the levels of serum ALT,AST,IL-1β,IL-6,and TNF-αand the expression of MPO in the liver tissues were lower in the point group than in the model group(P<0.05);there were no significant differences between the non-point group and the model group in comparing these indexes(P>0.05).Conclusion Preintervention with electroacupuncture at Zusanli(ST36)and Yanglingquan(GB34)can produce a certain protective effect on hepatic ischemia-reperfusion injury,playing a role in improving liver function,inhibiting inflammatory cytokine release,and reducing neutrophil infiltration.
