摘要
为了建立快捷的玫瑰冠等位基因分型方法,试验基于LAMP技术,设计特异性引物,在结合浊度法与金属离子指示剂法等方法下,对玫瑰冠等位基因分型进行可行性研究。结果显示:三组引物中,Rose1-1无特异性,Rose4-1和Rose4-2具有一定的特异性,其中Rose4-1在利用HNB作为染料,不添加环引物时的体系组合,可以在24个已知基因型的个体中准确筛选出纯合子与杂合子个体。结果表明,通过LAMP技术进行玫瑰冠个体基因分型具有可行性,同时也为优化其他性状的分子标记辅助选择提供了有益参考。
In order to establish a more convenient genotyping method of rose comb alleles,the study designed specific prim⁃ers based on LAMP(Loop-mediated isothermal amplification)technology,and carried out a feasibility study on rose comb geno⁃typing combined with turbidity method and metal ion indicator method.The results showed that among the three sets of primers,Rose1-1 had no specificity,while Rose4-1 and Rose4-2 had certain specificity.The system combination of Rose4-1 could accu⁃rately identify homozygous and heterozygous individuals among 24 chickens with known genotypes when using HNB as dye with⁃out adding a loop primer.The results indicated that genotyping of rose comb by lamp technology was feasible,which also provided a useful reference for optimizing molecular marker assisted selection of other traits.
