摘要
采用碱性蛋白酶酶解提取豆渣中不溶性膳食纤维,在单因素试验的基础上,采用响应面法优化提取工艺,并对其持水力、持油力和溶胀性进行测定。结果表明:最佳工艺条件为酶解pH 9.0、酶解温度51℃、酶解时间4.1 h、酶添加量8250 U/g,在此条件下豆渣中不溶性膳食纤维含量为91.35%;不溶性膳食纤维的持水力、持油力及溶胀性分别为8.79 g/g、7.96 g/g、14.99 mL/g,与豆渣相比,分别提高了76.51%、92.74%、173.04%。
The insoluble dietary fiber in soybean residue was extracted by alkaline protease enzymatic hydrolysis.On the basis of single factor test,the extraction process was optimized by response surface method,and its water holding capacity,oil holding capacity and swelling capacity were determined.The results showed that the optimum technological conditions were enzymatic hydrolysis pH 9.0,enzymatic hydrolysis temperature 51°C,enzymatic hydrolysis time 4.1 h,and amount of enzyme 8250 U/g.Under these conditions,the content of insoluble dietary fiber in soybean residue was 91.35%.The water holding capacity,oil holding capacity and swelling capacity of insoluble dietary fiber were 8.79 g/g,7.96 g/g and 14.99 mL/g,respectively,which were 76.51%,92.74%and 173.04%higher than that of soybean residue.
