山茱萸果核提取物不同洗脱部位对急性药物性肝损伤的作用研究

摘要 目的研究山茱萸果核提取物大孔树脂不同洗脱部位对对乙酰氨基酚(APAP)所致小鼠急性肝损伤(ALI)的保护作用。方法通过大孔吸附树脂将山茱萸果核提取物分别用0%、30%、60%和90%乙醇进行洗脱,得到4个洗脱部位(ESCOA、ESCOB、ESCOC、ESCOD)。再以生化检测和组织病理学结果,评价山茱萸果核不同洗脱部位的低、高剂量组(以生药量计为4.68 g·kg^(-1)、12.48 g·kg^(-1))对APAP所致小鼠ALI的保护作用。结果组织病理学观察结果表明,模型组小鼠的肝组织可见炎性细胞浸润,伴有肝细胞变性、片状坏死,而山茱萸果核ESCOA高剂量组、ESCOB低剂量组及ESCOC各剂量组对这些病变均有不同程度的改善;生化检测结果表明,与模型组相比,各组小鼠血清中谷丙转氨酶水平均显著降低,ESCOA低剂量组和ESCOC各剂量组小鼠血清中谷草转氨酶水平显著降低,各组小鼠肝脏组织中SOD水平均显著升高,ESCOA低剂量组、ESCOB高剂量组、ESCOC各剂量组小鼠肝脏组织中GSH-Px显著升高,ESCOA低剂量组及ESCOC、ESCOD各剂量组小鼠血清中MDA水平显著降低。结论山茱萸果核提取物的不同洗脱部位均对APAP所致小鼠ALI有一定的保护作用,其机制可能与抗氧化应激反应有关。 Objective To determine the protective effect of different ethanol elution parts of macroporous resin(MR)from the extract of seeds of Cornus officinalis Sieb.et Zucc.(ESCO)on acetaminophen(APAP)-induced acute liver injury(ALI)in mice.Methods The ESCO extract was isolated and further purified by the MR into four elution fractions(ESCOA,ESCOB,ESCOC,and ESCOD).Then,the hepatoprotective effect of different elution parts of ESOC extract in low-dose and high-dose groups(4.16and 12.48 g·kg^(-1),calculated by crude drug)on APAP-induced ALI mice model was evaluated by the serum index and histopathological examination.Results The histopathological examination showed that inflammatory cell infiltration was observed in the liver tissue of the model group,accompanied by hepatocyte degeneration and flaky necrosis.While the lesions of the liver tissue were relieved in each groups.The biochemical test showed that compared with the model group,the serum levels of glutamic-pyruvic in each group were decreased significantly;the serum levels of glutamic-oxaloacetic transaminase in ESCOA and ESCOC low-dose group and ESCOC high-dose group were decreased significantly;the liver levels of SOD in all groups were increased significantly;the liver levels of GSH-Px in ESCOA low-dose group,ESCOB high-dose group,and both ESCOC groups were increased;and the serum levels of MDA in ESCOA low-dose group,and both ESCOC and ESCOD groups were decreased.Conclusion Different elution parts of ESOC extract have protective effect against APAP-induced ALI in mouse model,whose mechanism may be related to the anti-oxidative stress reaction.
