摘要
目的:探讨赣南地区脑卒中患者及Hcy水平与MTHFR碱基位点C677T、A1298C突变、CBS基因碱基位点T833C、G919A突变的关联性。方法:选择赣南地区300例脑卒中患者(观察组)和300例健康体检者(对照组),运用全自动生化分析检测血浆中相关生化指标及Hcy水平,运用扩增阻碍突变系统法技术检测研究对象MTHFR基因C677T、A1298C多态性,CBS基因T833C、G919A多态性,分析MTHFR基因C677T、A1298C多态性,CBS基因T833C、G919A多态性与脑卒中及Hcy的相关性。结果:相比对照组,观察组的Hcy水平显著增加存在统计学差异(P<0.05);赣南地区比较两组的MTHFR碱基位点C677T、A1298C突变比较存在统计学差异(P<0.05);赣南地区比较两组的CBS基因碱基位点T833C、G919A突变比较存在统计学差异(P<0.05);相比对照组,观察组的MTHFR基因C677T、A1298C多态性,CBS基因T833C、G919A不同突变位点的Hcy水平均存在显著性差异(P<0.05)。Person相关性分析结果显示,赣南地区脑卒中患者Hcy与MTHFR、CBS基因不同分型均存在相关性(P<0.05)。结论:Hcy水平增加是脑卒中的风险因子,赣南地区脑卒中及Hcy水平与MTHFR碱基位点C677T、A1298C突变、CBS基因碱基位点T833C、G919A突变具有显著性差异,且存在相关性。
Objective:To explore the relationship between the level of Hcy and the mutations of MTHFR base sites C677T,A1298C,CBS gene base sites T833C,G919A in stroke patients in Gannan area.Methods:A total of 300 patients with stroke(observation group)and 300 healthy people(control group)in Gannan area were selected to use automatic biochemical analysis to detect the relevant biochemical indicators and Hcy level in plasma,and the amplification blocking mutation system technology was used to detect the MTHFR gene C677T,A1298C polymorphism,CBS gene T833C,G919A polymorphism,MTHFR gene C677T,A1298C polymorphism,CBS gene T833C The association of G919A polymorphism with stroke and Hcy.Results:Compared with the control group,the level of Hcy in the observation group increased significantly(P<0.05).The MTHFR base mutations of C677T and A1298C in the two groups were statistically different P;There was statistical difference between the two groups of CBS gene base sites T833C and G919A mutations in southern Jiangxi(P<0.05).Compared with the control group,the MTHFR gene C677T,A1298C polymorphism and the Hcy level of different mutation sites of CBS gene T833C and G919A in the observation group were significantly different(P<0.05).The results of Person correlation analysis showed that there was correlation between Hcy and different genotypes of MTHFR and CBS genes in stroke patients in Gannan area(P<0.05).Conclusion:The increase of Hcy level is a risk factor for stroke.There is significant difference and correlation between stroke and Hcy level and MTHFR base sites C677T,A1298C mutations,CBS gene base sites T833C,G919A mutations in southern Jiangxi.
