血清miR-28-5p、HMGB1表达与多发性骨髓瘤患者病理特征及预后的关系

摘要 目的探讨血清miR-28-5p、高迁移率族蛋白B1(HMGB1)表达与多发性骨髓瘤(MM)患者病理特征及预后的关系。方法选取2018年1月至2019年6月于河南省南阳市中心医院就诊的MM患者124例为病例组,选取同期健康志愿者50名为对照组,分析两组血清miR-28-5p、HMGB1表达水平与患者病理特征及其预后的关系。结果与对照组比较,病例组血清miR-28-5p表达水平降低、血清HMGB1水平升高,差异有统计学意义(t=30.327、16.269,P均<0.05);不同D-S分期、ISS分期患者随着分期越晚血清miR-28-5p表达水平降低,差异有统计学意义(F=19.541、22.173,P均<0.05);不同ISS分期患者随着分期越晚血清HMGB1水平越高,差异有统计学意义(F=6.891,P<0.05);存活患者miR-28-5p水平高于死亡患者、HMGB1水平低于死亡患者,差异有统计学意义(t=4.416、2.546,P<0.05)。高表达水平miR-28-5p、低水平HMGB1的累计生存率更高,差异有统计学意义(χ^(2)=16.930、12.417,P均<0.05)。结论MM患者血清miR-28-5p表达水平降低、HMGB1表达水平升高,两项指标均与患者预后有关,或可作为MM患者预后评价的血清生物学指标。 Objective To investigate the relationship between serum miR⁃28⁃5p and high mobility group box 1(HMGB1)expression and the pathological features and prognosis of multiple myeloma(MM)patients.Methods 124 MM patients who visited Nanyang Central Hospital of Henan Province from January 2018 to June 2019 were selected as the case group,and 50 healthy volunteers of the same period were selected as the control group.The relationship between the expression levels of serum miR⁃28⁃5p and HMGB1 and the pathological characteristics and prognosis of patients in the two groups was analyzed.Results Compared with the control group,serum mir⁃28⁃5p expression levels decreased and serum HMGB1 levels increased in the case group,the difference was statistically significant(t=30.327,16.269,P<0.05).The expression level of serum miR⁃28⁃5p in patients with different D⁃S stages and ISS stages decreased with the later stage,and the difference was statistically significant(F=19.541,22.173,P<0.05).Patients with different ISS stages had higher serum HMGB1 levels with the later stages,and the difference was statistically significant(F=6.891,P<0.05).The level of miR⁃28⁃5p in surviving patients was higher than that in dead patients,and the level of HMGB1 was lower than that in dead patients,and the difference was statistically significant(t=4.416,2.546,P<0.05).The cumulative survival rate was higher in patients with high expression level of mir⁃28⁃5p and low level of HMGB1,the difference was statistically significant(χ^(2)=16.930,12.417,P<0.05).Conclusion The expression level ofserum miR⁃28⁃5p decreased and the expression level of HMGB1 increased in patients with MM.Both indicators are related to the prognosis of patients,and may be used as serum biological indicators for the evaluation of the prognosis of MM patients.
