通过GEO数据库筛选宫颈癌发生的相关基因

摘要 目的利用数据库筛选宫颈癌致病的关键基因,探讨宫颈癌的发病机制。方法从高通量基因表达数据库(GEO)下载宫颈癌相关的芯片数据GSE89657、GSE63678和GSE29570,利用GEO2R在线分析软件筛选出宫颈癌差异基因748个,分别对差异基因进行基因本体富集分析(GO)、京都基因和基因组百科全书(KEGG)通路分析以及蛋白质-蛋白质相互作用网络(PPI)分析。结果3个数据库共呈现32个共同差异基因,分别为15个上调基因,17个下调基因。差异基因导入DAVID数据库发现与宫颈癌细胞凋亡和基因表达负调控、癌症通路有关。发现10个差异基因成为PPI网络复合体中的中枢基因,其表达水平的改变与宫颈癌的不良预后有关。其中拓扑异构体酶Ⅱa(TOP2A)在宫颈癌的诊断中最为重要。结论通过GEO数据库分析宫颈癌的基因芯片数据,获取与其预后密切相关的基因,为疾病的早期诊断提供分子水平上的参考依据。 Objective Using the database to screen the key genes of cervical cancer and explore the pathogenesis of cervical cancer.Methods The chip data GSE89657,GSE63678 and GSE29570 related to cervical cancer were downloaded from the Gene Expression Omnibus(GEO),and 748 differential genes of cervical cancer were screened out by GEO2R online analysis software.The differential genes were analyzed by gene ontology enrichment analysis(GO),Kyoto Encyclopedia of genes and genomes(KEGG)pathway analysis and protein-protein interaction network(PPI)analysis.Results There were 32 differential genes in the three databases,including 15 up-regulated genes and 17 down regulated genes.Differential genes were introduced into DAVID database and found to be related to cell apoptosis,negative regulation of gene expression and cancer pathways in cervical cancer.It was found that 10 differential genes became the central genes in the PPI network complex,and the change of their expression levels was related to the poor prognosis of cervical cancer.Among them,TopoisomeraseⅡalpha(TOP2A)was the most important in the diagnosis of cervical cancer.Conclusion GEO database was used to analyze the gene chip data of cervical cancer to obtain the genes closely related to its prognosis,so as to provide the reference basis for the early diagnosis of the disease at the molecular level.
