摘要
目的探讨羽扇豆醇通过上调FOXO1表达促进上皮性卵巢癌细胞凋亡的机制。方法设立对照组,紫杉醇组(500μmol/mL),羽扇豆醇低、高剂量组(25μmol/mL、50μmol/mL)。每孔设6个平行样,培养72 h。培养结束后,采用MTT法测定细胞增殖水平,Transwell室测定细胞侵袭水平,流式细胞仪测定细胞凋亡水平,RT-PCR法及蛋白印迹法测定细胞FOXO1 mRNA和蛋白水平。结果与对照组比较,紫杉醇组、羽扇豆醇低剂量组、羽扇豆醇高剂量组存活率、穿膜数降低(P<0.05);与紫杉醇组比较,羽扇豆醇低、高剂量组存活率、穿膜数升高(P<0.05);与羽扇豆醇低剂量组比较,羽扇豆醇高剂量组存活率、穿膜数降低(P<0.05)。与对照组比较,紫杉醇组、羽扇豆醇低剂量组、羽扇豆醇高剂量组凋亡率、FOXO1 mRNA和蛋白表达水平升高(P<0.05);与紫杉醇组比较,羽扇豆醇低、高剂量组凋亡率、FOXO1 mRNA和蛋白表达水平降低(P<0.05);与羽扇豆醇低剂量组比较,羽扇豆醇高剂量组凋亡率、FOXO1 mRNA和蛋白表达水平升高(P<0.05)。结论羽扇豆醇可促进上皮性卵巢癌HO8910细胞凋亡,其机制与羽扇豆醇可能上调上皮性卵巢癌HO8910细胞FOXO1 mRNA和蛋白表达有关。
Objective To explore the mechanism of lupeol promoting apoptosis of epithelial ovarian cancer cells by up-regulating the expression of FOXO1.Methods Control group,paclitaxel group(500μmol·mL–1),and lupeol low,high dose groups(25μmol·mL–1,50μmol·mL–1)were established.Six parallel samples were set in each group,and cultured for 72 h.After culture,MTT method was used to measure cell proliferation,Transwell chamber was used to measure cell invasion,flow cytometry was used to measure cell apoptosis,and RT-PCR and Western blotting were used to measure FOXO1 mRNA and protein levels.Results Compared with control group,the survival rate and membrane penetration number of paclitaxel group,lupeol low,high dose groups were decreased(P<0.05);compared with paclitaxel group,the survival rate and membrane penetration number of lupeol low and high dose groups were increased(P<0.05);compared with lupeol low dose group,the survival rate and membrane penetration number of lupeol high dose group were decreased(P<0.05).Compared with the control group,the apoptosis rate,FOXO1 mRNA and protein expression levels in paclitaxel group,lupeol low,high dose groups were increased(P<0.05);compared with paclitaxel group,the apoptosis rate,FOXO1 mRNA and protein expression levels in lupeol low,high dose groups were decreased(P<0.05);compared with lupeol low dose group,the apoptosis rate,FOXO1 mRNA and protein expression levels in lupeol high dose group were increased(P<0.05).Conclusion Lupeol can promote the apoptosis of epithelial ovarian cancer HO8910 cells,and its mechanism is related to that lupeol may up-regulate the expression of FOXO1mRNA and protein in epithelial ovarian cancer HO8910 cells.
