三叶青地上部分水溶性多糖的解热作用研究

摘要 目的:研究三叶青地上部分(THP)中多糖的化学性质及解热作用及机制。方法:采用DEAE法和凝胶渗透色谱法对三叶青地上部分水溶性粗多糖进行提取和分离。通过均匀性、分子量、单糖组成和FTIR分析对THP进行了表征。采用啤酒酵母诱导热疗试验检测THP的解热作用。结果:口服200和400 mg/kg的THP可明显抑制啤酒酵母诱导的小鼠体温升高,降低小鼠血清中细胞因子尤其是前列腺素E2(PGE2)的产生。在脂多糖(LPS)前预防性给予THP可降低高水平的细胞因子,表明THP的解热作用可能与LPS竞争,与toll样受体4(TLR4)相互作用,从而进一步调节细胞因子的分泌。结论:THP的抗炎可能与其通过控制细胞因子的分泌调节宿主免疫功能有关。 Objective:To study the chemical properties,antipyretic effect and mechanism of polysaccharides in the aerial parts of A.clover(THP).Methods:DEAE method and gel permeation chromatography were used to extract and separate the water-soluble crude polysaccharides from the aerial part of Clover.THP was characterized by homogeneity,molecular weight,monosaccharide composition and FTIR analysis.The antipyretic effect of THP was detected by brewer’s yeast induced hyperthermia test.Results:Oral administration of 200 and 400 mg/kg THP could significantly inhibit the increase in body temperature in mice induced by Saccharomyces cerevisiae,and reduce the production of cytokines,especially prostaglandin E2(PGE2)in mouse serum.Prophylactic administration of THP before lipopolysaccharide(LPS)reduced high levels of cytokines,suggesting that the antipyretic effect of THP may compete with LPS to interact with toll-like receptor 4(TLR4),thereby further regulating cytokine secretion.Conclusion:The anti-inflammation of THP may be related to its regulation of host immune function by controlling the secretion of cytokines.
