摘要
Nod样受体蛋白3(nucleotide-binding domain-like receptor 3,NLRP3)是炎症小体中关键的调控蛋白之一,参与机体炎症反应的调控,并与痛风、关节炎、Ⅱ型糖尿病和癌症等疾病的产生发展紧密相关。近年来有研究表明NLRP3相关通路调控的炎症反应参与药物毒性作用发生发展过程。研究NLRP3通路在药物毒性作用中的机制有利于为临床安全有效用药提供指导,同时对于减轻药物不良反应具有重要意义。因此,拟对NLRP3相关信号通路作用机制、NLRP3通路相关药物毒性作用机制及NLRP3通路相关药物减毒研究三个方面进行总结。
Nucleotide-binding domain(NOD)-like receptor protein 3(NLRP3)is one of the crucial regulatory proteins in NLRP3-inflammasome,which is involved in the regulation of inflammatory response in the body.NLRP3 is associated with gen⁃eration and development of various diseases,for example,gout,arthritis,type II diabetes and cancer.Toxic and side effects of drugs cause acute or chronic inflammation which cause cell stress and tissue injury.The study of the mechanism of NLRP3 path⁃way in drug toxicity made for the guidance for safe and valid drug use in clinical practice,which is of great significance for reduc⁃ing drug toxicity and side effects.The review contains mechanism of NLRP3 related signaling pathway and mechanism of drug toxicity about NLRP3 and researches of reducing drug toxicity via NLRP3 pathway.
