马铃薯NCED基因家族全基因组鉴定与表达分析

摘要 脱落酸(ABA)在植物响应生物胁迫和非生物胁迫过程中具有重要调节作用。9-顺式-环氧类胡萝卜素双加氧酶(NCED)是ABA合成限速酶,位于ABA生物合成通路上游。为明确马铃薯NCED基因家族成员以及NCED基因表达特性,本研究在全基因组水平鉴定马铃薯NCED基因家族成员,并通过染色体定位、理化性质表征、系统发育树构建、基因结构分析以及启动子顺式作用元件鉴定对该基因家族成员进行系统研究。基于马铃薯参考基因组,共鉴定到10个马铃薯NCED基因,染色体定位分析显示这些基因分布在4条染色体上;基因理化性质分析表明,该基因家族蛋白分子量介于50~70 kD之间,等电点介于5.48~8.71之间;启动子序列分析显示该基因家族具有大量与多种植物激素相关的顺式作用元件;最后通过实时荧光定量PCR明确接种茄链格孢(Alternaria solani)后马铃薯NCED基因家族的表达模式。本研究为深入挖掘NCED基因家族在马铃薯与茄链格孢互作中的作用提供了理论基础。 Abscisic acid(ABA)plays important regulatory roles in plant response to biotic and abiotic stresses.9-cis-epoxy carotenoid dioxygenases(NCEDs)is rate-limiting enzyme in ABA biosynthesis and locate at the upstream of the ABA biosynthesis pathway.In this study,we identified the potato NCED gene family at whole genome level,and systematically studied its members by chromosome mapping,physicochemical properties characterization,phylogenetic construction,gene structure analysis and cis-acting element identification in order to classify the NCED gene family and verify the expression pattern of NCED gene.Based on potato reference genome,we identified 10 NCED genes which were located on 4 chromosomes respectively.Physicochemical properties characterization showed that the molecular weight of NCEDs were between 50~70 kD and the isoelectric point of the 10 proteins were from 5.48~8.71.Promoter sequence analysis showed that the gene family contained multiple plant hormone-related elements.The transcriptional expression patterns of NCED gene family in potato were determined by real-time fluorescence quantitative PCR analysis post Alternaria solani inoculation This study provides fundamental evidences for further exploring potato and Alternaria solani interaction.
