摘要
为研究筛选出牛源肝素钠氧化的最优工艺参数,对成品的质量进行检测,并对其结构进行分析。采用正交试验法,对牛源肝素钠氧化步骤的工艺参数进行优化筛选,并对关键指标进行检测,再对二糖组成进行分析。结果表明,牛源肝素钠工艺氧化步骤的最佳反应条件为反应温度40±2℃,pH值9.0±0.2,双氧水加量2.5%,反应时间6 h。按最优氧化条件,该步骤的脱色率为94.6%,效价收率为97.3%,二糖组成表明牛源肝素与猪源肝素在结构上有所差异,该工艺稳定且高效。
In order to select the optimal process parameters for the oxidation of bovine heparin,and the quality of the product would have tested and its structure would have analyzed.Orthogonal test was used to optimize the technological parameters of the oxidation step of bovine heparin sodium,and then to detect the key indicators and disaccharide composition.The result showed that:The optimum reaction conditions for the oxidation of bovine heparin were:The reaction temperature 40±2℃,the pH value 9.0±0.2,the amount of hydrogen peroxide 2.5%,and the reaction time 6 hours.According to the optimal oxidation conditions,the decolorization rate of this step was 94.6%,and the rate of recovery was 97.3%,and the composition of disaccharide showed that bovine heparin was structurally different from porcine heparin.The process was stable and efficient.
