蒙药齿缘草及混淆品的化学轮廓及镇痛抗炎作用比较研究

Comparative Study on the Chemical Profile,Analgesic and Anti-inflammatory Effects of Mongolian Medicine Eritrichium Rupestre and Its Confusions

目的:建立蒙药齿缘草的HPLC对照指纹图谱,比较齿缘草及其混淆品(东北齿缘草、板蓝根、大青叶)的化学轮廓;通过小鼠镇痛抗炎实验,评价齿缘草及其混淆品的镇痛抗炎作用,为齿缘草与其混淆品替代使用提供参考依据。方法:HPLC法建立齿缘草的对照指纹图谱,流动相为乙腈-0.4%磷酸水溶液(梯度洗脱),流速设定为1.0 mL·min^(-1),检测波长选择270 nm,柱温设置为30℃,进样量为10μL,采用标准对照法对齿缘草的共有峰进行识别;混淆品(东北齿缘草、板蓝根、大青叶)化学轮廓色谱图分别在上述相同的色谱条件下获得,再应用“中药色谱指纹图谱相似度评价系统”软件评价混淆品与齿缘草对照图谱的相似度。分别通过小鼠热板、醋酸扭体和耳廓肿胀实验,比较蒙药齿缘草及其混淆品东北齿缘草、板蓝根和大青叶的抗炎镇痛作用。结果:建立蒙药齿缘草的对照指纹图谱,标定17个共有峰,并指认出1、3、6、17号峰对应的化合物分别为绿原酸、咖啡酸、芦丁和槲皮素;东北齿缘草、板蓝根、大青叶与齿缘草对照指纹图谱的相似度分别为0.850、0.558和0.300,其中,东北齿缘草与齿缘草对照指纹图谱有14个共有峰,板蓝根有13个共有峰,大青叶有5个共有峰。除大青叶外,齿缘草、东北齿缘草和板蓝根均表现出较强的镇痛抗炎作用。结论:本研究初步认为齿缘草与其混淆品东北齿缘草、板蓝根的替代使用具有一定的合理性,有必要开展深入研究。

Objective:To establish the HPLC control fingerprint of Mongolian medicine Eritrichium rupestre,to compare the chemical profiles of Eritrichium rupestre and its confusions(Eritrichum mandshuricum,Radix isati-dis and Folium isatidis),and to study the analgesic and anti-inflammatory effects of Eritrichium rupestre and its confusions through analgesic and anti-inflammatory experiments in mice,which provide reference data for the substitute use of Eritrichium rupestre and its confusions.Methods:The HPLC method was used to establish the control fingerprint of Eritrichium rupestre.Acetonitrile-0.4%phosphoric acid aqueous solution was used as the mobile phase(gradient elution),the flow rate was 1.0 mL·min^(-1),detection wavelength was 270 nm,column temperature was 30℃,and injection volume was 10 jjlL.The standard control method was used to identify the com mon peaks of Eritrichium rupestre.Under the same chromatographic conditions,the chemical profile chromatograms of Eritrichum mandshuricum,Radix isatidis and Folium isatidis were measured respectively,and the similarity evaluation was conducted with the"Similarity evaluation system of traditional Chinese medicine chromatographic fingerpriirt".The anti-inflammatory and analgesic effects of Mongolian medicine Eritrichium rupestre and its confusions such as Eritrichum mandshuricum,Radix isatidis and Folium isatidis were compared by hot plate 9 acetic acid writhing and auricle swelling experiments in mice.Results:The control fingerprint of Mongolian medicine Eritrichium rupestre was established,and 17 common peaks and 4 corresponding compounds(chlorogenic acid,caHeic acid,rutin and quercetin)of peaks 1,3,6 and 17 were identified.Compared with the control fingerprint of Eritrichium rupestre,the similarities of fingerprint of Eritrichum,mandshuricum,Radix isatidis and Folium isatidis were 0.850,0.558 and 0.300 respectively.Compared with the control fingerprint of Eritrichium rupestre,Eritrichum mandshuricum has 14 common peaks 9 Radix isatidis has 13 common peaks and Folium isatidis has 5 common peaks.Execpt Folium isatidis,Eritrichium rupestre,Eritrichum mandshuricum and Radix isatidis showed strong analgesic and anti-inflammatory effects.Conclusion:This study preliminarily concluded that the alternative use of Mongolian medicine Eritrichium rupestre and its confusions(Eritrichum,mandshuricum , Radix isatidis)is reasonable,and further research is necessary.
