高尤-13对CUMS大鼠血清SP、VIP和抑郁行为的影响

Effects of Gaoyou-13 on Serum SP,VIP and Depressive Behavior in CUMS Rats

目的:观察高尤-13对慢性不可预知温和应激(CUMS)大鼠血清P物质(substance P,SP)、血管活性肠肽(vasoactive intestinal peptide,VIP)和抑郁行为的影响。方法:将48只雄性SD大鼠按体重随机分为正常组、模型组、氟西汀组、高尤-13低、中、高剂量组,采用慢性不可预知温和应激与孤养结合的方法制备抑郁模型大鼠,正常组、模型组灌胃蒸馏水,氟西汀组按3.3 mg·kg^(-1)灌胃氟西汀,高尤-13低、中、高剂量组分别按0.25、0.50、1.00 g·kg^(-1)灌胃高尤-13,以体质量、旷场实验、糖水消耗实验评价行为学变化,用ELISA法测定大鼠血清SP和VIP含量。结果:体重,与正常组比较,模型组21 d、28 d、35 d、42 d体重下降;与模型组比较,高尤-13低剂量组28 d、35 d,高尤-13中剂量组28 d,高尤-13高剂量组21 d、28 d、35 d、42 d,氟西汀组28 d体重增加,差异有统计学意义(P<0.05)。旷场试验,与正常组相比,模型组21 d、42 d水平跨格运动次数和直立运动次数减少;与模型组比较,高尤-13低、中、高剂量组21 d、42 d水平跨格运动次数升高,高尤-13低、中、高剂量组21 d直立运动次数升高,差异有统计学意义(P<0.05)。糖水消耗实验,与正常组比较,模型组21 d、42 d糖水消耗量降低;与模型组比较,高尤-13中剂量组21 d、42 d,高尤-13高剂量组42 d,氟西汀组21 d、42 d糖水消耗量升高,差异有统计学意义(P<0.05)。大鼠血清SP、VIP含量,与正常组比较,模型组血清SP含量升高;与模型组比较,高尤-13高剂量组、氟西汀组血清SP含量降低,差异有统计学意义(P<0.05)。结论:高尤-13抗抑郁作用与调节血清SP、VIP相关。

Objective:To observe the effects of Gaoyou-13 on serum substance P(SP),vasoactive intestinal peptide(VIP)and depressive behavior in chronic unpredictable mild stress(CUMS)rats.Methods:Forty-eight male SD rats were randomly divided into six groups:a normal group,a model group,a Fluoxetine group,Gaoyou-13 low,medium,and high dose groups according to body weight.Rat depression model was established by the combination of CUMS and solitary rearing.The normal group and model group were given distilled water intragas-trically,the Fluoxetine group was given Fluoxetine at 3.3 mg·kg^(-1),the Gaoyou-13 low-dose,medium-dose and high-dose groups were intragastrically given Gaoyou-13 at 0.25,0.5 and 1.0 g·kg^(-1),respectively.The behavioral changes were evaluated by body mass,open field test and sucrose consumption test,and the contents of SP and VIP in rat serum were determined by ELISA.Results:Compared with the normal group,the body weight of model group decreased on 21 d,28 d,35 d and 42 d;Compared with the model group the body weight of the Gaoyou-13 low-dose group increased on 28 d and 35 d,of the Gaoyou-13 medium-dose group increased on 28 d,of the Gaoyou-13 high-dose group increased on 21 d,28 d,35 d and 42 d,of the Fluoxetine group increased on 28 d,and the differences were statistically significant(P<0.05).In the open field test,compared with the normal group,the number of horizontal cross-grid movements and vertical movements were reduced in the model group on 21 d and 42 d;Compared with the model group,the number of horizontal cross-grid movements was increased in the Gaoyou-13 low,medium and high dose groups on 21 d and 42 d,and the number of vertical movements was increased in the Gaoyou-13 low,medium and high dose groups on 21 d,and the differences were statistically significant(P<0.05).In the sugar water consumption experiment,compared with the nonnal group,the sugar water consumption in the model group decreased on 21 d and 42 d;Compared with the model group,the sugar water consumption in the Gaoyou-13 medium dose group increased on 21 d and 42 d,in the Gaoyou-13 high dose group increased on 42 d,and in the Fluoxetine group increased on 21 d and 42 d,and the differences were statistically significant(P<0.05).In terms of serum SP and VIP contents in rats,compared with the normal group,the serum SP in the model group increased;Compared with the model group,the serum SP levels decreased in the Gaoyou-13 high dose group and Fluoxetine group,and there were statistical difference(P<0.05).Conclusion:The antidepressant effect of Gaoyou-13 is correlated with the regulation of serum SP and VIP.
