细枝叶下珠愈伤组织诱导影响因素研究

摘要 [目的]筛选出较优的细枝叶下珠愈伤组织诱导培养基配方,提高湿生植物细枝叶下珠的繁殖系数,满足日益增长的海绵城市发展要求。[方法]利用单因素控制变量法进行组织培养,以细枝叶下珠的无芽和一节芽茎段为外植体,MS培养基和B5培养基为基本培养基,6-BA、IBA为外源激素,设置3个激素浓度配比处理:6-BA 1.0 mg/L+IBA 0.2 mg/L;6-BA 1.5 mg/L+IBA 0.2 mg/L;6-BA 1.5 mg/L+IBA 0.1 mg/L。[结果 ]75%酒精和0.2%次氯酸钠消毒后不同外植体的存活率相同,不同培养基的存活率也相同,均为62.5%;一节茎段诱导率比不带芽茎段高出73.3个百分点,MS培养诱导率比B5培养基高出28.3个百分点;细枝叶下珠在MS+6-BA 1.0 mg/L+IBA 0.2 mg/L培养基上愈伤组织诱导效果较好,诱导率达53.8%。[结论]筛选出适宜培养基,为工厂化育苗提供参考。 [Objective]The aim was to screen out the optimal callus induction medium formula of Phyllanthus leptoclados,and improve the reproduction coefficient of hygrophyte P.leptoclados,and meet the growing requirements of sponge city development.[Method]The tissue culture was carried out by single factor control variable method.The stem segments without bud and with bud were used as explants.MS medium and B5 medium were basic media.6-BA and IBA were exogenous hormones.Set 3 hormone concentration ratio treatment:6-BA 1.0 mg/L+IBA 0.2 mg/L;6-BA 1.5 mg/L+IBA 0.2 mg/L;6-BA 1.5 mg/L+IBA 0.1 mg/L.[Result]The survival rate of different explants was the same after disinfected by 75%alcohol and 0.2%sodium hypochlorite,and the survival rate of different media was the same,62.5%.The induction rate of one stem segment was 73.3%higher than that of stem segment without bud,and the induction rate of MS culture was 28.3%higher than that of B5 medium.The callus induction rate of P.leptoclados on MS+6-BA 1.0 mg/L+IBA 0.2 mg/L medium was 53.8%.[Conclusion]The suitable medium was screened to provide reference for factory seedling cultivation.
