基于TCGA数据库的肺腺癌G2/M检查点基因预后模型的建立与应用

摘要 目的基于癌症基因组图谱(TCGA)数据库建立肺腺癌(LUAD)的G2/M检查点相关基因风险预后模型。方法通过TCGA数据库获取并整合了LUAD基因表达数据及临床数据,根据Reactome定义的G2/M检查点相关基因集筛选出影响预后的G2/M检查点相关基因,通过单因素和多因素COX分析建立风险预后模型,对模型高、低风险组进行生物学标志基因集和KEGG通路富集的差异分析并进行免疫相关性分析。结果预后模型由7个基因组成,该模型生存预测性能的AUC值为0.726。高风险组的总体生存率明显低于低风险组(P<0.05)。细胞增殖的相关通路在高风险组中富集。风险评分的增高与多种免疫细胞的浸润比例存在一定相关性(P<0.05)。结论由7个基因组成的风险预测模型可作为独立预测因子来有效预测LUAD的预后。 Objective To establish a risk prognostic model of G2/M checkpoint-related genes in lung adenocarcinoma(LUAD)based on The Cancer Genome Atlas(TCGA)database.Methods The gene expression data and clinical data of lung adenocarcinoma were obtained and integrated through the TCGA database,and G2/M checkpoint-related genes affecting prognosis were screened according to the G2/M checkpoint-related gene set defined by Reactome.The risk prognostic model was established by univariate and multivariate COX analysis,and the differential analysis of biological marker gene set and KEGG pathway enrichment were performed for the high and low risk groups of the model,and the immune correlation was analyzed.Results The prognostic model consisted of 7 genes,and the AUC value of the model′s survival prediction performance was 0.726.The overall survival rate of the high-risk group was significantly lower than that of the low-risk group(P<0.05).Pathways associated with cell proliferation were enriched in high-risk groups.There was a certain correlation between the increase of risk score and the infiltration ratio of various immune cells(P<0.05).Conclusion The prognostic model composed of 7 genes can be used as an independent prognostic predictor to predict the prognosis of LUAD.
