葛根素对DHF大鼠心肌细胞形态和心肌组织肌联蛋白表达的影响

摘要 目的 研究葛根素(Pue)对舒张性心力衰竭(DHF)大鼠心肌细胞形态和心肌组织肌联蛋白表达的影响。方法 Wistar大鼠采用腹主动脉缩窄法建立DHF模型,术后4周,分为模型组,Pue高、中、低剂量组(180、120、80 mg/kg)4组(n=10),另有假手术组10只。在给药4周后,通过16导生理记录仪测量所得的血流动力学指标评估模型建立成功,应用透射电子显微镜观察心肌细胞微观结构,应用免疫组化法(SV)测定心肌组织肌联蛋白表达。结果 电镜下心肌细胞形态观察结果 :Pue组与空白组比较,Pue高剂量组、模型组心肌细胞肥大,胶原纤维明显增多、增粗及排列紊乱,细胞间距增大,间质增生,心肌间隙有大量胶原纤维堆积,Pue低剂量及中剂量组细胞微观结构差异相对较小;Pue组与模型组比较,Pue中剂量组、Pue低剂量组细胞排列紧密,整齐有序,间质增生不明显,Pue高剂量组细胞微观结构并无明显差异。免疫组化法测定心肌组织肌联蛋白表达:模型组与空白组相比,心肌细胞肌联蛋白表达显著下降(P <0.05)。Pue组与模型组比较,Pue高剂量组心肌细胞肌联蛋白表达无明显变化;Pue中剂量组心肌细胞肌联蛋白表达显著增强(P <0.05);低剂量组心肌细胞肌联蛋白表达明显增强(P <0.05)。结论 Pue中剂量、Pue低剂量可能通过改善DHF大鼠血流动力学指标,改善DHF大鼠心肌细胞、组织微观结构,提高titin的表达,而发挥抗心室重构的作用,缓解舒张功能障碍,逆转DHF的进程。此外,高剂量的Pue不能被用于DHF的治疗。 Objective To study the Effects of different dose of puerarin(Pue)morphology of cardimyocytes and expression titin of diastole heart failure rat model.Methods Diastole heart failure model was established by the coarctation of abdominal aorta.Four weeks after operation,forty diastole heart failure rats were divided into four groups ran domly as follows,model(normal saline 2 mL),puerarin(180 mg/kg),puerarin(120 mg/kg),puerarin(80 mg/kg)with 10 rats for each group and 10 sham operation rats was taken as control(normal saline,2 mL).After 4 weeks administration,the microstructures of cardiomyocytes were observed by transmission electron microscopy,cardiac function was determined by catheter recording instrument.the expression of titin was examined by immunohistochemical method.Results Observation results of myocardial cells under the electron microscope,compared with sham operation grocup,in pue high-dose group and model grocup myocardial cells were hypertrophy,collagenousfibers increased、thickened and disordered,cell spacing increased、interstitium hyperplasia,and a large number of collagenousfibers were accumulated in the space between myocardium,the differences in cell microstructure between pue low-dose and pue medium-dose groups were relatively small.Compared with the model group,the cells in pue middle-dose group and pue low-dose group were closely arranged and orderly,and the interstitial hyperplasia was not obvious.There was no significant difference in the cell microstructure in pue high-dose group.Immunohistochemical method was used to determine the expression of titin in myocardial tissue,compared with the sham group,the expression of titin in model group was significantly decreased(P<0.05).Compared with model group,the expression of titin in Pue high-dose group had no significant change.The expression in Pue medium-dose group was significantly increased(P<0.05),the expression of titin in cardiomyocytes in low-dose group was significantly increased(P<0.05).Conclusion Medium dose of pue and low dose of pue may play an anti-ventricular remodeling effect,alleviate diastolic dysfunction and reverse the process of DHF by improving the hemodynamic indexes of DHF rats,improving the microstructure of cardiomyocytes and tissues in DHF rats,and improving the expression of titin.In addition,high doses of pue cannot be used for DHF treatment.
