摘要
目的:优化大黄牡丹颗粒的提取工艺。方法:采用高效液相色谱法,以丹皮酚、大黄酸、大黄素、大黄酚及固体总量为评价指标,考察提取次数、提取时间、加水量对提取工艺的影响,基于熵权法分析各指标权重系数,结合Box-Behnken响应面法结果优化大黄牡丹颗粒提取工艺。结果:丹皮酚、大黄酸、大黄素、大黄酚质量浓度分别为1.55~64.50、5.43~226.41、1.20~50.00、1.26~52.70μg·mL^(–1)时与峰面积呈良好的线性关系。采用熵权法进行综合评分,丹皮酚、大黄酸、大黄素、大黄酚及固体总量的权重系数(Wj)分别为0.1147、0.3225、0.2477、0.2078、0.1073,且根据Box-Behnken响应面法优化的结果,确定大黄牡丹颗粒最佳提取工艺为提取次数为2次、提取时间为30 min、加水量为10倍。结论:优化后的大黄牡丹颗粒提取工艺稳定性高、重复性好,适用于批量生产。
Objective:To optimize the extraction process of Dahuang Mudan Granules.Methods:With paeonol,rhein,emodin,chrysophanol and total solids as evaluation indexes,high performance liquid chromatography(HPLC)was used to investigate the extraction times,extraction time and water addition.The weight coefficient of each index was analyzed based on entropy weight method,and the extraction parameters of Dahuang Mudan Granules were optimized by combination with Box-Behnken response surface methodology.Results:Paeonol,rhein,emodin and chrysophanol showed good linear relationships at 1.55-64.50,5.43-226.41,1.20-50.00,1.26-52.70μg·mL^(–1),respectively.In the comprehensive evaluation by entropy weight method,the weight coefficients(Wj)of paeonol,rhein,emodin,chrysophanol and total solids were 0.1147,0.3225,0.2477,0.2078 and 0.1073,respectively.According to Box-Behnken response surface methodology,the optimum extraction process of Dahuang Mudan Granules was determined as follows:extraction times 2,extraction time 30 min,and water addition 10 times.Conclusion:The optimized extraction process of Dahuang Mudan Granules has high stability and good repeatability,which is suitable for mass production.
