骨碎补总黄酮对去势小鼠海马神经元细胞的抗凋亡作用

摘要 目的:研究骨碎补总黄酮(Drynaria fortunei,DF)对卵巢摘除所致认知功能障碍小鼠海马神经元的保护作用。方法:本研究将90只昆明小鼠随机平均分为空白组、模型组、假手术组(SHAM组)、雌二醇阳性药(E2)组、DF组、DF+ICI182780(阻断剂)组,每组15只。采用新物体识别实验检测小鼠的记忆能力;应用HE染色、透射电镜观察小鼠海马区的细胞形态;通过TUNEL法测定小鼠的神经元细胞凋亡比。结果:与空白组相比较,模型组小鼠的识别指数明显下降,海马区神经元细胞数量降低,神经元细胞凋亡比明显增大;与模型组相比较,SHAM组、E2组及DF组小鼠的识别指数均上升,但后者在统计学中具有更为显著的意义,且SHAM组、E2组和DF组小鼠神经元细胞数量较多、细胞结构清晰可见,神经元细胞凋亡比明显降低;与DF组相比较,DF+ICI182780组细胞凋亡比明显升高,神经元细胞数量明显下降。结论:DF可以改善去势雌性小鼠的认知功能障碍,抑制海马神经元细胞凋亡,这可能是通过介导雌激素受体(Estrogen receptor,ER)实现的。 Objective:To study the protective effect of total flavonoids of Rhizoma Drynariae(Drynaria fortunei,DF)on hippocampal neurons in mice with cognitive dysfunction induced by ovariectomy.Methods:The 90 Kunming mice were randomly divided into control group,model group,sham surgery group(SHAM group),estradiol-positive drug(E2)group,DF group,or DF+ICI182780(blocker)group,and there were 15 mice each group.To know the memory ability of experimental mice in this study,new object recognition experiment was employed.Morphology of mouse hippocampal zone was visualized by HE staining or using transmission electron microscopy.The neuronal apoptosis ratio of mice was determined through TUNEL.Results:Compared to control group,the recognition performance was significantly downregulated,the neuronal cells in the hippocampus were decreased,and the neuronal apoptosis ratio was significantly increased in the model group;Compared to Model group,the recognition performances in SHAM group,E2 group or DF group were upregulated.However only the latter was with significance in statistics.There were more neuronal cells in SHAM group,E2 group and DF group than those in Model group.Also,the cellular structure was clearly visible,and the apoptotic ratio of neuronal cells was significantly decreased in the same groups.Compared to DF group,the apoptotic ratio in DF+ICI182780 group was significantly increased while the neuronal cells were significantly downregulated.Conclusion:DF could improve cognitive dysfunction and inhibit the apoptosis of hippocampal neurons in castrated female mice,which may work through ER.
