摘要
目的研究去整联蛋白金属蛋白酶家族癸蛋白1(a disintegrin and metalloprotease like decysin 1,ADAMDEC1)对T细胞急性淋巴细胞白血病(T-cell acute lymphoblastic leukemia,T-ALL)细胞生物学行为的影响及其潜在的分子机制。方法qRT-RCR和Western blot检测ADAMDEC1在人T淋巴细胞H9与T-ALL细胞Jurkat和CCRF-CEM的表达。CCK-8法检测ADAMDEC1siRNA干扰表达后T-ALL细胞增殖的变化。流式细胞术检测ADAMDEC1siRNA干扰后T-ALL细胞凋亡情况。Western blot检测干扰ADAMDEC1后凋亡及Wnt/β-catenin通路相关分子蛋白水平的变化。结果ADAMDEC1在CCRF-CEM和Jurkat细胞中高表达。干扰ADAMDEC1后CCRFCEM和Jurkat细胞增殖能力降低,细胞凋亡率增高。干扰ADAMDEC1可促进GSK-3β的表达,抑制β-catenin、p-GSK-3β和c-Myc的蛋白水平。Wnt/β-catenin通路激活剂CHIR-99021可部分地逆转干扰ADAMDEC1对T-ALL细胞增殖和凋亡的影响。结论干扰ADAMDEC1后通过抑制Wnt/β-catenin通路,进而抑制T-ALL细胞增殖,诱导细胞凋亡。
Objective To study the effect of a disintegrin and metalloprotease like decysin 1(ADAMDEC1)on the biological function of T-cell acute lymphoblastic leukemia(T-ALL)cell and its potential molecular mechanism.Methods The expression of ADAMDEC1 in human T lymphocyte H9 and T-ALL cells Jurkat and CCRF-CEM were detected by qRT-RCR and Western blot.CCK-8 assay was used to detect the proliferation of T-ALL cells after ADAMDEC1 siRNA interference expression.Flow cytometry was used to detect the apoptosis of T-ALL cells after ADAMDEC1 siRNA interference expression.Western blot was used to detect the changes of apoptosis and Wnt/β-catenin pathway-related molecular protein expression levels after interfering with ADAMDEC1.Results ADAMDEC1 was highly expressed in CCRF-CEM and Jurkat cells.After interfering with ADAMDEC1,the proliferation ability of CCRF-CEM and Jurkat cells decreased and the apoptosis rate increased.Interfering with ADAMDEC1 can promote the expression of GSK-3βand inhibit the protein expression levels ofβ-catenin,p-GSK-3βand c-Myc.Wnt/β-catenin pathway activator CHIR-99021 could partially reverse the effect of ADAMDEC1 interference on the proliferation and apoptosis of T-ALL cells.Conclusion Knockdown of ADAMDEC1 inhibits Wnt/β-catenin pathway to inhibit T-ALL cell proliferation and induce apoptosis.
