铁死亡在动脉粥样硬化中的作用及其靶向治疗研究进展

Research advances in role of ferroptosis in atherosclerosis and its targeted therapy

动脉粥样硬化(atherosclerosis,AS)是一种以脂质代谢异常为特征的慢性炎症性疾病,其导致的心脑血管疾病有着较高致残和致死率。铁死亡是一种新型的细胞死亡形式,它依赖铁的调控,触发多个抗氧化系统,由脂质过氧化激活,参与调节AS、泡沫细胞形成、内皮功能障碍、斑块形成和斑块内出血等病变过程,因此靶向铁死亡有望成为治疗AS及预防并发症的新方向。本文综述了铁死亡作用机制,重点讨论了铁死亡与AS之间的联系,并归纳总结了近年来靶向铁死亡治疗AS的相关研究进展,以期为AS的治疗提供理论依据。

Atherosclerosis(AS)is a chronic inflammatory disease characterized by abnormal lipid metabolism,resulting in cardiovascular and cerebrovascular diseases with high disability and fatality.Ferroptosis is a new form of cell death,which depends on the regulation of iron and triggers multiple antioxidant systems,it is activated by lipid peroxidation,and participates in the regulation of AS,foam cell formation,endothelial dysfunction,plaque formation,intra-plaque hemorrhage and other pathological processes.Therefore,targeting ferroptosis is expected to become a new direction for the treatment of AS and the prevention of complications.This paper reviews the mechanism of ferroptosis,focuses on the relationship between ferroptosis and AS,and summarizes the research progress of targeted ferroptosis in the treatment of AS in recent years to provide theoretical reference for the treatment of AS.
