摘要
目的临床观察发现闽产荔枝草对冻疮具有一定疗效,黄酮是其主要成分,本研究旨在优化荔枝草总黄酮提取工艺,探讨其防治冻疮作用,为其临床应用提供科学依据。方法建立闽产荔枝草总黄酮紫外-可见分光光度检测方法,进行单因素试验,并采用响应面Box-Behnken设计优化荔枝草总黄酮提取工艺;建立大鼠冻疮模型,观察荔枝草总黄酮对冻疮大鼠肿胀程度、皮损修复的影响。结果所建立荔枝草总黄酮紫外-可见分光光度法方法学考察符合要求,表明该方法稳定可靠;响应面优化后得到提取工艺条件为:70%乙醇,超声时间40 min,液料比40∶1,在此条件下,荔枝草总黄酮提取量达(9.85±0.05)mg·g^(-1)。荔枝草总黄酮提取液对冻疮大鼠皮损情况、肿胀率均有良好治疗作用。结论荔枝草总黄酮检测方法稳定可靠,优化后的提取条件简便易行,适用于荔枝草总黄酮提取;该提取条件下荔枝草总黄酮对大鼠冻疮有一定疗效。
Objective Preliminary clinical observation shows that Salvia plebeia produced in Fujian has a certain curative effect on chilblain,and flavonoids are its main active component.This study aims to optimize the extraction process of total flavonoids from Salvia plebeia,explore its role in preventing and treating chilblain,and provide scientific basis for its clinical application.Methods UV-VIS spectrophotometric method for the determination of total flavonoids from Salvia plebeia(TFSP)produced in Fujian was established,a single factor experiment was carried out,and the extraction process of TFSP was optimized by response surface box Behnken design;The rat model of chilblain was established to observe the effects of TFSP on the swelling degree and skin lesion repair of chilblain rats.Results The established UV-VIS spectrophotometry for TFSP met the requirements,indicating that the method was stable and reliable;The extraction conditions were as follows:70%ethanol,ultrasonic time 40 min,liquid-solid ratio 40∶1.Under these conditions,the extraction amount of TFSP was(9.85±0.05)mg·g^(-1).TFSP had a good therapeutic effect on the skin lesions and swelling rate of rats.Conclusion The detection method of TFSP was stable and reliable,and the optimized extraction conditions were simple and feasible,which was suitable for the extraction of TFSP;Under this extraction condition,TFSP had a certain effect on frostbite in rats.
