HPLC-MS/MS分析麻口皮子药内生真菌培养基条件下次生代谢产物与原药材

HPLC-MS/MS Analysis of Secondary Metabolites from Zanthoxylum simulans Endophytic Fungi Cultured in Different Media and Active Pharmaceutical Ingredient

摘要 采用液质联用技术(HPLC-MS/MS)分析麻口皮子药内生真菌在不同培养基条件下的次生代谢产物,并与麻口皮子药原药材进行比较。HPLC指纹图谱显示,13种培养基条件下的次生代谢产物与原药材具有一定的相似性,其中J2、D8培养基条件下的次生代谢产物与原药材具有多个共有峰。总离子流(TIC)图谱显示,13种培养基条件下的次生代谢产物与原药材有3个相同化合物,分子量分别为565(t_(R)=50.2 min)、679(t_(R)=52.3 min)、792(t_(R)=53.5 min),这3个化合物均未见文献报道,数据库中也无相关数据,推测为新化合物;其它培养基条件下的次生代谢产物之间成分各有异同,通过二级质谱及裂解规律,推导出了部分可能的化合物及其分子式。表明,可通过选择合适的培养基条件定向调控特定有效组分的合成,为提高麻口皮子药活性成分含量提供了一种新途径。 We analyzed the secondary metabolites from Zanthoxylum simulans endophytic fungi cultured in different media by HPLC-MS/MS method,and compared with active pharmaceutical ingredient(API).The results of HPLC fingerprint spectra show that the secondary metabolites cultured in 13 media have certain similarity with the API,and the secondary metabolites cultured in J2 and D8 media have many common peaks with the API.The results of TIC spectra show that the secondary metabolites cultured in 13 media have three same compounds as the API,with molecular weights of 565(t_(R)=50.2 min),679(t_(R)=52.3 min),and 792(t_(R)=53.5 min),respectively.None of these three compounds has been reported in the literature,and there is no relevant data in the database,so they are presumed to be new compounds.The components of secondary metabolites cultured in other media have similarities and differences.Some possible compounds and their molecular formulas have been deduced by secondary mass spectrometry and pyrolysis law.The results indicate that the synthesis of specific effective components can be controlled by selecting appropriate medium,which provides a new way to improve the content of active components of Zanthoxylum simulans.
