通窍活血汤治疗缺血性脑卒中的疗效观察及对ET-1、CGRP因子影响

Observation on Therapeutic Effect of Tongqiao Huoxue Decoction(通窍活血汤)on Ischemic Stroke and Its Effect on ET-1 and CGRP Factors

目的通过动物实验,探讨通窍活血汤治疗缺血性脑卒中的临床疗效及对ET-1、CGRP因子的影响。方法实验以48只健康雄性SD大鼠为对象,其中8只大鼠作为假手术组,其余40只用于制备MCAO动物模型。将造模成功的大鼠随机分为4组,模型组及通窍活血汤高、中、低剂量组,每组各8只(排除死亡和不符合标准的模型)。假手术组与模型组灌服同体积的生理盐水,通窍活血汤组按高、中、低剂量灌胃,分别持续灌服14 d,1次/d,给药第14天测定大鼠神经功能情况,并于末次给药后2 h处死。用RT-qPCR法检测大鼠脑组织ET-1及CGRP含量,用TTC染色法观察大鼠梗死情况。结果与假手术组相比,模型组与通窍活血汤各剂量组血清ET-1表达水平均有所上升(P<0.05),脑组织CGRP表达水平有差异(P<0.05),呈下降趋势,有统计学意义;与模型组相比,大鼠血清ET-1含量在通窍活血汤各剂量组的表达水平下降(P<0.05),脑组织CGRP表达水平升高(P<0.05),有统计学意义;与通窍活血汤高剂量组相比,大鼠血清ET-1表达在通窍活血汤中、低剂量组升高(P<0.05),脑组织CGRP在通窍活血汤中、低剂量组表达降低(P<0.05),有统计学意义,说明通窍活血汤高剂量组比中、低剂量组疗效更优。结论通窍活血汤治疗急性缺血性脑卒中可能是通过影响血管内皮因子的舒缩功效,发挥对血管内皮功能的保护作用。

Objective To explore the clinical efficacy of Tongqiao Huoxue Decoction(通窍活血汤)in the treatment of ischemic stroke and its effect on ET-1 and CGRP factors through animal experiments.Methods 48 healthy male SD rats were selected as subjects,of which 8 rats were used as sham operation group,and the other 40 rats were used to establish MCAO animal model.The successful rats were randomly divided into 4 groups:model group and Tongqiao Huoxue Decoction high,middle and low dose groups,8 rats in each group(excluding death and substandard models).The rats in the sham operation group and the model group were given the same volume of normal saline,and the rats in the Tongqiao Huoxue Decoction group were given intragastric administration at high,middle and low doses for 14 days,once a day,respectively.The neurological function of the rats was measured on the 14th day,and the rats were killed 2 hours after the last administration.The contents of ET-1 and CGRP in rat brain tissue were detected by RTqPCR method,and the infarct was observed by TTC staining.Results Compared with the sham operation group,the expression level of serum ET-1 in model group and Tongqiao Huoxue Decoction group increased(P<0.05),and the expression level of CGRP in brain tissue decreased(P<0.05).Compared with the model group,the expression level of ET-1 in serum decreased(P<0.05)and the expression level of CGRP in brain tissue increased in Tongqiao Huoxue Decoction groups(P<0.05).Compared with the high dose group of Tongqiao Huoxue Decoction,the expression of serum ET-1 increased in the middle and low dose groups of Tongqiao Huoxue Decoction(P<0.05),while the expression of CGRP in brain tissue decreased in the middle and low dose groups of Tongqiao Huoxue Decoction(P<0.05).Conclusion Tongqiao Huoxue Decoction may protect vascular endothelial function by affecting the relaxing and contractile effect of vascular endothelial factor in the treatment of acute ischemic stroke.
